Restriction Enzyme Mapping

Cleave plasmid DNA and map the relative positions of restriction sites.

In this experiment, a plasmid DNA is cleaved with different combinations of restriction enzymes. By determining the fragment size and using agarose gel electrophoresis, the relative positions of the restriction sites can be mapped.

Additional items needed but not included: DNA Electrophoresis equipment, 5 to 50 µl micropipettes, Water Bath, Transilluminator (UV or White-Light depending on which stain you use), Microwave or Hot plate.
