A vitamin/enzyme cofactor found in milk, eggs, malted barley, liver, kidney, heart, leafy vegetables. Richest natural source is yeast. Occurs in the free form only in the retina of the eye, in whey, and in urine; bioactive forms occurring in tissues and cells are riboflavin monophosphate and flavine-adenine dinucleotide. Used as a photoinitiator for polymerization of polyacrylamide gels. It forms free radicals in aqueous solution in the presence of light. Riboflavin photodecomposes to leucoflavin. No free radicals are formed in the absence of oxygen but traces of oxygen allows for leucoflavin to reoxidize with free-radical generation. The catalysts TEMED or DMAPN are commonly added to speed up the free radical formation. The free radicals will cause acrylamide and bis-acrylamide to polymerize to form a gel matrix which can be used for electrophoresis. Riboflavin is commonly used in the stacking gel for non-denaturing polyacrylamide electrophoresis because native proteins can be senstive to persulfate ions from ammonium persulfate. Another advantage of riboflavin over ammonium persulfate is that it will not start polymerization until the gel is illuminated. Riboflavin causes less denaturing than other initiators. Gel strength can be improved when used with PTO (Pyrithione). Soluble in 0.1 M Sodium Hydroxide (50 mg/ml - clear, yellowish-orange solution); only slightly soluble in water.

Triton® X-100 is an alkylaryl polyether alcohol. It is a nonionic surfactant prepared by the reaction of octylphenol with ethylene oxide. The number ("-100") relates only indirectly to the number of ethylene oxide units in the structure.
Triton X-100 is a non-ionic surfactant and emulsifier which is often used in biochemical applications to solubilize proteins. Triton X-100 has no antimicrobial properties. It is considered as a comparatively mild detergent, non-denaturing, and is reported in numerous references as a routinely added reagent. It does absorb in the ultraviolet region of the spectrum, however, so can interfere with protein quantitation.
The principal uses of Triton® X-100 is in industrial and household detergent applications and in emulsifying agents. It is employed in almost every type of liquid, paste, and powdered cleaning compound, ranging from heavy-duty industrial products to gentle detergents for fine fabrics. It is also important ingredient of primary emulsifier mixtures used in the manufacture of emulsion polymers, stabilizers in latex polymers, and emulsifiers for agricultural emulsion concentrates, and wettable powders.

No significant inhibition against PLKs 1 to 3 is observed for CFI-400945 at a concentration of 50 μM. CFI-400945 can attenuate the growth of breast cell line, as well as other tumor cell lines significantly. CFI-400945 selectively inhibits PLK4 in cells, but also has certain activity against AURKB, TRKA, TRKB and Tie2/TEK (only 10 kinases showed more than 50% inhibition among 290 kinases). The cytokinesis failure and subsequent polyploidization by CFI-400945 treatment indicate that the cell death in cancer cell lines is at least partly achieved through inhibition of AURKB. No significant inhibition is observed for PLKs 1 to 3 (IC50 s > 50 μM) probably due to the most divergent structure of PLK4 compared to other polo-like kinases 1 to 3. Cancer cells treated with CFI-400945 exhibit effects consistent with PLK4 kinase inhibition, including dysregulated centriole duplication, mitotic defects, and cell death.

Citric acid can form complexes with various cations, particularly with iron and calcium. In animals, citric acid improves the utilization of nutritional calcium. Citric acid is produced commercially by fermentation of carbohydrates derived from corn starch and from beet molasses.
Citric Acid, Trisodium Salt, Dihydrate is used as a substrate for citrate lyase, a buffer component; an anticoagulant. For anticoagulation use it is typically used at a concentration of approximately 0.129 M (i.e. for 4.5 mL blood use 16.0 mg sodium citrate and 2.1 mg citric acid).
To make a sodium citrate buffer use equimolar concentrations (typically approximately 0.05 M concentration) of citric acid, free acid and sodium citrate. Add equal volumes of each solution and titrate to the desired pH.

Folic acid (Vitamin B9 and folate) is essential to numerous bodily functions. Folic acid (FA) and dihydrofolic acid (FAH2) are substrates of dihydrofolate reductase(s) which reduce them to tetrahydrofolate (THF), which in turn supports ‘one carbon’ transfer. Tetrahydrofolates are required for de novo synthesis of purines, thymidylic acid and various amino acids and for post-translational methylation (epigenetics).

β-NAD, a pyridine nucleotide and biologically active form of nicotinic acid, is a coenzyme necessary for the catalytic reaction of certain enzymes. It occurs in living cells primarily in the oxidized state. Serves as a coenzyme of the dehydrogenases, especially in the dehydrogenation of primary and secondary alcohols. NAD usually acts as a hydrogen acceptor, forming NADH which then serves as a hydrogen donor in the respiratory chain.
Many metabolites and enzymes of biological interest are present in tissues at low concentrations. With the use of β-NAD as a catalyst intermediate and several enzymes in a multistep system, known as enzyme cycling, much greater sensitivity for detection of these components is achieved. The reduced form, β-NADH, is fluorescent whereas β-NAD is not. This difference in fluorescence provides a sensitive fluorescent measurement of the oxidized or reduced pyridine nucleotides at concentrations down to 10-7 M.
Electron acceptor. β-NAD is a carrier for hydride ion, forming b-NADH. Hydride ion is enzymatically removed from a substrate molecule by the action of dehydrogenases such as malic dehydrogenase and lactic dehydrogenase. Such enzymes catalyze the reversible transfer of a hydride ion from malate or lactate to b-NAD to form the reduced product, b-NADH. Unlike b-NAD which has no absorbance at 340 nm, b-NADH absorbs at 340 nm (EmM = 6.22).
