Anti-FAF1 Rabbit Polyclonal Antibody

Anti-FAF1 Rabbit Polyclonal Antibody

Supplier: Rockland Immunochemical

Fas-associated protein 1 (FAF1) was initially identified as a Fas-binding pro-apoptotic protein that is component of the death-inducing signaling complex in Fas-mediated apoptosis. FAF1 can also induce apoptosis in the absence of extrinsic death signals when overexpressed although it does not contain typical death motifs such as the death domain, death effector domain, and caspase recruitment domain. Overexpression of FAF1 also decreases the basal level of NF-kB activity in transfected 293 cells, inhibits NF-kB activity induced by TNF-a, IL-1b and lipopolysaccharide, and prevents NF-kB translocation to the nucleus, suggesting that another role of FAF1 is to negatively regulate the activity of NF-kB. FAF1 can also interact with the inflammatory signaling PYRIN-containing Apaf-1-like proteins (PYPAFs, also called NALPs) such as PYPAF1, PYPAF2 (NALP2), and PYPAF7, suggesting FAF1 may also be involved in the inflammation pathway. Multiple differentially spliced isoforms of FAF1 are known to exist.

Anti-EBI3 Rabbit Polyclonal Antibody

Anti-EBI3 Rabbit Polyclonal Antibody

Supplier: Rockland Immunochemical

EBI3 is a subunit in two distinct heterodimeric cytokines: interleukin-27 (IL-27) and IL-35. Like interleukin-23 (IL-23), IL-27 is a recently discovered member of the IL-6/IL-12 family of proinflammatory and immunoregulatory cytokines. It exists as a heterodimer composed of the p40-related protein EBI3 and an IL-12 p35-related protein termed p28. IL-27 is produced after activation by antigen-presenting cells and induces proliferation of naïve but not memory CD4+ T-cells. It acts by binding to its receptor WSX-1 (also known as TCCR) and gp130 which results in the activation of a Jak/STAT signaling cascade, suggesting the IL-27 is involved in the regulation of immune processes. It has been suggested that IL-27 can also be used as a therapeutic agent against cancer as it can also induce tumor-specific anti-tumor activity mediated through CD8+ T-cells, IFN-gamma, and T-bet. IL-35 is composed of EBI3 and the p35 subunit of IL-12 and has been reported to have therapeutic effects against collagen-induced arthritis by expanding the population of regulatory T cells and suppressing Th17 cells. At least two isoform of EBI3 are known to exist.

Anti-IgG Rabbit Polyclonal Antibody (DL488)

Anti-IgG Rabbit Polyclonal Antibody (DL488)

Supplier: Rockland Immunochemical

Conjugated Anti-Monkey IgG (H&L) DyLight™ 488 antibody generated in rabbit detects specifically monkey IgG heavy and light chains. Secreted as part of the adaptive immune response by plasma B cells, immunoglobulin G constitutes 75% of serum immunoglobulins. IgG binds to viruses, bacteria, as well as fungi and facilitates their destruction or neutralization via agglutination (and thereby immobilizing them), activation of the compliment cascade, and opsinization for phagocytosis. The whole IgG molecule possesses both the F(c) region, recognized by high-affinity Fc receptor proteins, as well as the F(ab) region possessing the epitope-recognition site. Both heavy and light chains of the antibody molecule are present. This DyLight™488 conjugated anti-Monkey IgG (H&L) secondary antibody is ideal for investigators who routinely perform immunofluorescence, flow cytometry, and more general immunoassays. When choosing a secondary antibody product, consideration must be given to species and immunoglobulin specificity, conjugate type, fragment and chain specificity, level of cross-reactivity, and host-species source and fragment. The emission spectra for this DyLight™ conjugate match the principle output wavelengths of most common fluorescence instrumentation.

Mouse Recombinant IL35 (from CHO Cells)

Supplier: Adipogen

Interleukin-35 (IL-35) is a novel IL-12 family cytokine produced by regulatory T cells (Treg) but not by resting or activated effector T cells (Teff). IL-35 is a heterodimeric protein composed of EBI3 (Epstein-Barr-Virus-induced gene 3) and IL-12a (p35). EBI3 is a downstream target of Foxp3, a transcription factor required for Treg-cell development and function, and thus Treg-cell restriction of IL35 occurs. Regulatory T cells are essential for maintaining self tolerance and preventing autoimmunity, and IL-35 is identified as a molecule that mediates the immune suppression function of Treg-cell. As an inhibitory cytokine, IL-35 induces proliferation of Treg-cell populations but suppresses Th17 cell development. Studies in mice show the absence of either IL-35 chain from Treg-cell reduces the cells' ability to suppress inflammation using an experimental model for inflammatory bowel disease. IL-35 is suggested as a potential target of immunotherapy. Recently, insufficient IL-35 levels were shown to play a pivotal role in the development of type 1 diabetes (T1D) and autoimmune diseases.

Human Recombinant IL-1alpha (from E. coli)

Supplier: Adipogen

The most prominent members of the interleukin-1 (IL-1) superfamily are IL-1alpha and IL-1beta. They lack a signal peptide and are secreted by an unconventional, endoplasmic reticulum-Golgi-independent mechanism. IL-1alpha was reported to be more widely and constitutively expressed and has intracellular functions, but also acts locally in a membrane-bound form by activating IL-1R1. Additionally, passive release of IL-1alpha upon cell death can trigger a sterile inflammatory response to dying cells. The cleavage of IL-1alpha is not mediated by caspase-1 and is not required for binding to IL-1R1. Recently it has been observed that all activators of the inflammasome NLRP3/NALP3 induce the simultaneous secretion of IL-1alpha and IL-1beta. Although most activators fully rely on the inflammasome for IL-1alpha secretion, some induce the processing and secretion of IL-1alpha in an inflammasome-independent manner.

Human Recombinant CD274 (from CHO Cells)

Supplier: Adipogen

Programmed death ligand 1 (PD-L1, B7-H1 or CD274) is a member of the growing B7 family of immune proteins that provide signals for both stimulating and inhibiting T cell activation. CD274 has been identified as one of two ligands for programmed death 1 (PD-1), a member of the CD28 family of immunoreceptors. CD274 is widely expressed in several organs such as heart, skeletal muscle, placenta and lung, and in lower amounts in thymus, spleen, kidney and liver. CD274 expression is upregulated in a small fraction of activated T and B cells and a much larger fraction of activated monocytes. CD274 expression is also induced in dendritic cells and keratinocytes after IFN-gamma stimulation. CD274 expression is also upregulated in a variety of tumor cell lines. Interaction of CD274 with PD-1 results in inhibition of TCR mediated proliferation and cytokine production, suggesting an inhibitory role in regulating immune responses. The CD274 - PD-1 pathway is involved in the negative regulation of some immune responses and may play an important role in the regulation of peripheral tolerance.

Human Recombinant TIM3 (from CHO Cells)

Supplier: Adipogen

The TIM (T cell/transmembrane, immunoglobulin and mucin) family plays a critical role in regulating immune responses, including allergy, asthma, transplant tolerance, autoimmunity and the response to viral infections. The unique structure of TIM immunoglobulin variable region domains allows highly specific recognition of phosphatidylserine (PtdSer), exposed on the surface of apoptotic cells. Tim-3, a type I transmembrane protein, contains an immunoglobulin and a mucin-like domain in its extracellular portion and a tyrosine phosphorylation motif in its cytoplasmic portion. TIM-3 is preferentially expressed on Th1 and Tc1 cells, and generates an inhibitory signal resulting in apoptosis of Th1 and Tc1 cells. TIM-3 is also expressed on some dendritic cells and can mediate phagocytosis of apoptotic cells and cross-presentation of antigen. Tim-3 functions to inhibit aggressive Th1-mediated auto- and alloimmune responses. Tim-3 pathway blockade by administration of Tim-3:Fc fusion protein accelerates diabetes in nonobese diabetic mice, causes hyperproliferation of Th1 cells and Th1 cytokine release in an experimental autoimmune encephalomyelitis (EAE) model and prevents acquisition of transplantation tolerance induced by costimulation blockade.

Human Recombinant Beclin-1 (from E. coli)

Supplier: Adipogen

Beclin-1 (Bcl-2 interacting coiled-coil protein; ATG6) is a multifunctional protein, well-known as a key regulator of autophagy and required for the initiation step in autophagosome formation. It interacts with several cofactors (Atg14L, UVRAG, Bif-1, rubicon, Ambra1, HMGB1, nPIST, VMP1, SLAM, IP3R, PINK and survivin) to regulate the lipid kinase PI3K protein and promote formation of PIK3C3/VPS34-containing complexes. These interactions induce autophagy, which is a process of programmed cell survival increased during periods of cell stress and extinguished during the cell cycle. Furthermore, beclin-1 is implicated in numerous biological processes, including adaptation to stress, development, endocytosis, cytokinesis, immunity, infection, tumorigenesis, aging and cell death. Beclin-1 is tightly regulated by diverse mechanisms, such as epigenetic silencing, microRNA regulation, post-translational modifications and protein-protein interactions. Multiple diseases are associated with deficiency or malfunction of beclin-1, including cancer and neurodegeneration.
