Adhesion to extracellular matrix regulates cell survival through both integrin engagement and appropriate cell spreading. Anoikis is the molecular mechanism of apop-tosis induced by integrin detachment. Bit1 (Bcl-2 inhibitor of transcription 1) was recently identified as being involved in this process. Bit1 is a mitochondrial protein that is released into the cytoplasm upon onset of apoptosis where it forms a complex with AES, a small Groucho/transducin-like enhancer of split (TLE) protein and induces caspase-independent apoptosis. Both AES and TLE proteins are transcriptional co-repressors that play important roles in neurogenesis, segmentation, and sex determination. It has been suggested that Bit1-AES complexes turn off a survival-promoting gene transcription program controlled by TLE. Interestingly, apoptosis of cells transfected with Bit1 and AES could be inhibited if the cells were allowed to attach to fibronectin through the alpha5beta1 integrin suggesting that the Bit1-AES pathway contributing to anoikis is regulated by integrins, and in particular, the alpha5beta1 integrin.

Fibrillarin Antibody detects Nop1p which was originally identified as a nucleolar protein of bakers yeast, Saccharomyces cerevisiae. The Nop1p protein is 327 amino acids in size (34.5kDa), is essential for yeast viability, and is localized in the nucleoli. The systematic name for S. cerevisiae Nop1 is YDL014W, and it is now known to be part of the small subunit processome complex, involved in the processing of pre-18S ribosomal RNA. Nop1p is the yeast homologue of a protein found in all eukaryotes and archea generally called fibrillarin. Fibrillarin/Nop1p is extraordinarily conserved, so that the yeast and human proteins are 67% identical, and the human protein can functionally replace the yeast protein. Patients with the autoimmune disease scleroderma often have strong circulating autoantibodies to a ~34kDa protein which was subsequently found to be fibrillarin. Recent studies show that knock-out of the fibrillarin gene in mice results in embryonic lethality, although mice with only one functional fibrillarin/Nop1p gene were viable. Cochlin is an extracellular matrix protein expressed in the cochlea and vestibule of the inner ear. Mutations in the cochlin gene are linked to the autosomal-dominant DFNA9 syndrome, a disorder characterized by hearing loss. Cochlin is also expressed in follicular dendritic cells of the spleen and the lymph nodes. The N-terminus of cochlin contains a LCCL domain also present in Factor C, a serine protease of the horseshoe crab Limulus, involved in activating the coagulation cascade in response to LPS. A recent study showed that in response to inflammation, LPS or bacterial infection, cochlin is cleaved by the proteases aggrecanases and secreted into blood circulation. The secreted cochlin (LCCL domain) seems to have a role in local cytokine production, recruitment of immune effector cells and bacterial clearance. Cochlin is secreted as two forms p8 and p18, whereas p18 being the glycosylated form of p8.

In humans, betatrophin is found to be predominantly expressed in the liver. Betatrophin levels are reduced by fasting and are elevated upon insulin resistance and during pregnancy. Betatrophin, according to preliminary data could bind to the macrophage receptor Marco and also to RTN4R, a neuronal receptor. Recently, a study using ANGPTL8 KO mice showed that ANGPTL8/Betatrophin does not play a role in beta cell proliferation nor in glycemic control as previously thought, but regulates plasma triglyceride levels.
