Periostin is a 90-kDa matricellular protein that consists of a typical signal sequence, followed by a cysteine-rich region, an EMI domain (protein-protein interactions), four tandem fasciclin-like domains that are responsible for integrin binding, and a C-terminal region. Periostin was originally isolated as an osteoblast-specific factor that functions as a cell adhesion molecule for pre-osteoblasts and in osteoblast recruitment, attachment and spreading. Periostin is also involved in many fundamental biological processes such as cell proliferation, cell invasion and angiogenesis. Periostin expression is increased by both transforming growth factor beta1 (TGF-beta1) and bone morphogenetic protein 2 (BMP-2). Changes in periostin expression are commonly detected in various cancers and pre-cancerous conditions, and periostin may be involved in regulating cancer cell activities that contribute to tumorigenesis, cancer progression and metastasis. Periostin has shown to be involved in many aspects of allergic inflammation, such as eosinophil recruitment, airway remodeling, development of a Th2 phenotype and increased expression of inflammatory mediators. It is evaluated as a biomarker for bronchial asthma and airway inflammation.

CD152 and CD28 are structurally homologous molecules that are members of the immunoglobulin (Ig) gene superfamily. Both CD152 and CD28 are composed of a single Ig V-like extracellular domain, a transmembrane domain and an intracellular domain. CD152 and CD28 are both expressed on the cell surface as disulfide-linked homodimers or as monomers. CD152 was originally identified as a gene that was specifically expressed by cytotoxic T lymphocytes. However, CD152 transcripts have since been found in both Th1 and Th2, and CD4+ and CD8+ T cell clones. Whereas, CD28 expression is constitutive on the surfaces of 95% of CD4+ T cells and 50% of CD8+ T cells and is down regulated upon T cell activation, CD152 expression is upregulated rapidly following T cell activation and peaks approximately 24 hours following activation. Although both CD152 and CD28 can bind to the same ligands, CD152 binds to B71 and B72 with 20-100-fold higher affinity than CD28.

Betatrophin (RIFL; Lipasin; Angiopoietin-like protein 8 (ANGPTL8)) is a newly discovered secreted protein of 198 aa that was proposed to promote beta cell proliferation and improve glucose tolerance in mice. Betatrophin may also function in inhibition of lipase activity and on serum triglyceride regulation. Betatrophin is expressed in the liver and in white and brown adipose tissue of mice. In humans, betatrophin is found to be predominantly expressed in the liver. Betatrophin levels are reduced by fasting and are elevated upon insulin resistance and during pregnancy. Betatrophin, according to preliminary data could bind to the macrophage receptor Marco and also to RTN4R, a neuronal receptor. Interleukin-21 (IL-21) is a key factor in the transition between innate and adaptive immune responses secreted by activated T cells. The IL-21 receptor (IL-21R) is expressed in lymphoid tissue, in particular by NK, B, T and dendritic cells, macrophages and endothelial cells. Recent evidence suggests that IL-21 plays a supportive role in the proliferation of T and B cells and influences the cytolytic activity of natural killer cells. IL-21 has been shown to up-regulate genes associated with innate immunity and to inhibit the differentiation of naïve T helper cells. IL-21 specifically inhibits IFN-gamma production from developing TH1 cells and is preferentially expressed by TH2 cells. IL-21/IL-21R interactions have a unique role in sequentially activating both innate and adaptive immune responses against poorly immunogenic tumors, leading to tumor rejection that is perforin dependent but IFN-gamma independent.

CCL2, also called monocyte chemotactic protein-1 (MCP-1), is a member of the C-C or beta chemokine family that is best known as a chemotactic agent for mononuclear cells. Fibroblasts, glioma cells, smooth muscle cells, endothelial cells, lymphocytes and mononuclear phagocytes can produce CCL2 either constitutively or upon mitogenic stimulation, but monocytes and macrophages appear to be the major source. In addition to its chemotactic activity, CCL2 induces enzyme and cytokine release by monocytes, NK cells and lymphocytes, and histamine release by basophils that express its receptor CCR2. Additionally, it promotes Th2 polarization in CD4+ T cells. The human CD134 protein is expressed only on activated CD4+ T blasts, and its ligand has been identified as gp34. Interactions between CD134 and its ligand in vivo are necessary for the differentiation of activated B cells into highly immunoglobulin-producing cells, however not involved in other pathways of antigen-driven differentiation of B cells such as development of memory cells in the germinal centers. Fibroblast growth factors (FGFs) constitute a family of heparin-binding polypeptides involved in the regulation of biological responses such as growth, differentiation and angiogenesis. The biological effects of FGFs are mediated by four structurally related receptor tyrosine kinases denoted FGFR1, FGFR2, FGFR3 and FGFR4. FGF-1 is a powerful mitogen of cells of mesodermal, ectodermal and endodermal origin. FGF-1 association with heparan sulfate is a prerequisite for activation of FGF receptors. FGF-1 plays a role in various stages of development and morphogenesis as well as in angiogenesis and wound healing processes. Recent data indicate a role of FGF-1 in inflammation and obesity. FGF-1 is selectively induced in fat cells by high-fat diet feeding and established the PPARgamma-FGF-1 axis as a critical pathway that regulates adipose tissue remodeling.
