Anti-8-oxo-dG Mouse Monoclonal Antibody [clone: N45.1]
Supplier: Adipogen
8-hydroxy-2'-deoxyguanosine (8-OHdG) is a modified base that occurs in DNA due to attack by hydroxyl radicals (incl. singlet oxygen and direct photodynamic action) that are formed as byproducts and intermediates of aerobic metabolism and during oxidative stress. There is increasing evidence to support the involvement of free radical reactions in the damage of biomolecules that eventually lead to several diseases in humans, such as atherosclerosis, cerebral and heart ischemia-reperfusion injury, cancer, rheumatoid arthritis, inflammation, diabetes, aging, and neurodegenerative conditions, such as Alzheimer’s disease. 8-OHdG is popular as a sensitive, stable and integral marker of oxidative damage in cellular DNA and can be detected in tissue, serum, urine and other biomaterials.

Human Recombinant IL35 (from CHO Cells)
Supplier: Adipogen
Interleukin-35 (IL-35) is a novel IL-12 family cytokine produced by regulatory T cells (Treg) but not by resting or activated effector T cells (Teff). IL-35 is a heterodimeric protein composed of EBI3 (Epstein-Barr-Virus-induced gene 3) and IL-12a (p35). EBI3 is a downstream target of Foxp3, a transcription factor required for Treg-cell development and function, and thus Treg-cell restriction of IL35 occurs. Regulatory T cells are essential for maintaining self tolerance and preventing autoimmunity, and IL-35 is identified as a molecule that mediates the immune suppression function of Treg-cell. As an inhibitory cytokine, IL-35 induces proliferation of Treg-cell populations but suppresses Th17 cell development. Studies in mice show the absence of either IL-35 chain from Treg-cell reduces the cells' ability to suppress inflammation using an experimental model for inflammatory bowel disease. IL-35 is suggested as a potential target of immunotherapy.

Mouse Recombinant B7-H4 (from CHO cells)
Supplier: Adipogen
B7-H4 is a B7 family member that negatively regulates T cell immunity by inhibiting of T cell proliferation, cytokine production and cell cycle progression. In vitro, B7-H4 inhibits CD4+ and CD8+ T cell proliferation, cytokine production and generation of alloreactive cytotoxic T cells (CTLs). In vivo, blockade of endogenous B7-H4 by specific monoclonal antibody promotes T cell responses. B7-H4 ia an important negative regulator of innate immunity through growth inhibition of neutrophils. B7-H4 is expressed on some tumor cancer cells. The role of B7-H4 in tumor progression may be to transform precancerous cells and then protect them from immunosurveillance.

Human Recombinant HES1 (from E. coli)
Supplier: Adipogen
bHLH proteins are essential for neurogenesis, myogenesis, hematopoiesis and sex determination. HES1 (Hairy and Enhancer of Split 1) is a basic Helix-Loop-Helix (bHLH) transcriptional repressor regulated by the Notch pathway and expressed in various tissues. HES1 is well known in development of different tissues, particularly endoderm derived endocrine organs like pancreas and neuroendocrine cells of lung. HES1 controls normal development by regulating proliferation and differentiation and so cell fate. Notch signaling plays both oncogenic and tumor suppressor roles depending on cell type. In contrast to T cell acute lymphoblastic leukemia (ALL), where Notch activation promotes leukemogenesis, induction of Notch signaling in B cell ALL (B-ALL) leads to growth arrest and apoptosis. The Notch target HES1 is sufficient to reproduce this tumor suppressor phenotype in B-ALL by interacting and activating PARP-1 and regulating proapoptotic signals.

Anti-CASP8 Monoclonal Antibody [clone: C15]
Supplier: Adipogen
Procaspase-8 belongs to the family of caspases. Binding of FasL to Fas leads to formation of a receptor complex at the cellular membrane, which was named DISC. The DISC consists of oligomerized receptors, the DD-containing adaptor molecule FADD, procaspase-8, procaspase-10 and c-FLIP. The DISC structure provides a platform for the oligomerization of procaspase-8 that allows two procaspase-8 homodimers to be in the close proximity leading to the initial activation of procaspase-8. At the first cleavage step, the N-terminal p43/p41 and the C-terminal p30 cleavage products are generated. Importantly, these cleavage products already possess catalytic activity. At the second cleavage step, p43/p41 and p30 are processed to p10 and p18, respectively, which leads to the generation of the active caspase-8 heterotetramer (p18/p10)2.

Anti-Nampt Rabbit Polyclonal Antibody
Supplier: Adipogen
Nicotinamide phosphoribosyltransferase (Nampt; pre-B cell colony-enhancing factor; PBEF; Visfatin) is an 52kDa adipokine secreted by adipose tissue and involved in the biosynthesis of nicotinamide adenine dinucleotide (NAD+). Two forms of Nampt exist, an intracellular form (iNampt) and an extracellular form (eNampt). While the function of iNampt as an essential and rate-limiting NAD+ biosynthetic enzyme is well established, the physiological role of eNampt is still a matter of debate. Nampt has various functions, including the promotion of vascular smooth muscle cell maturation and inhibition of neutrophil apoptosis. It activates insulin receptor and has insulin-mimetic effects, lowering blood glucose and improving insulin sensitivity. The protein is highly expressed in visceral fat and serum levels of the protein correlate with obesity.

Mouse Recombinant CD279 (from CHO cells)
Supplier: Adipogen
CD279 (Programmed Cell Death Protein 1; PD-1) is a type I transmembrane protein belonging to the CD28/CTLA-4 family of immunoreceptors that mediate signals for regulating immune responses. Members of the CD28/CTLA-4 family have been shown to either promote T cell activation (CD28 and ICOS) or downregulate T cell activation (CTLA-4 and PD-1). CD279 is expressed on activated T cells, B cells, myeloid cells and on a subset of thymocytes. In vitro, ligation of CD279 inhibits TCR-mediated T cell proliferation and production of IL-1, IL-4, IL-10 and IFN-gamma. In addition, CD279 ligation also inhibits BCR mediated signaling. CD279 deficient mice have a defect in peripheral tolerance and spontaneously develop autoimmune diseases.

Anti-CASP11 Monoclonal Antibody [clone: 4E11]
Supplier: Adipogen
Caspase-11 not only activates caspase-1, that is required for the maturation of proinflammatory cytokines such as interleukin-1 (IL-1) and interleukin-18 (IL-18), but also activates caspase-3, leading to cellular apoptosis under pathological conditions. In most cells, caspase-11 is only expressed upon induction with pro-inflammatory stimuli. Caspase-11 is an essential mediator of endotoxic shock and caspase-11-deficient mice are resistant to endotoxic shock. LPS-induced caspase-11 regulates lymphocyte apoptosis by activating both caspase-3 and caspase-7.

Mouse Recombinant IL21 (from CHO Cells)
Supplier: Adipogen
Interleukin-21 (IL-21) is a key factor in the transition between innate and adaptive immune responses secreted by activated T cells. The IL-21 receptor (IL-21R) is expressed in lymphoid tissue, in particular by NK, B, T and dendritic cells, macrophages and endothelial cells. Recent evidence suggests that IL-21 plays a supportive role in the proliferation of T and B cells and influences the cytolytic activity of natural killer cells. IL-21 has been shown to up-regulate genes associated with innate immunity and to inhibit the differentiation of naïve T helper cells. IL-21 specifically inhibits IFN-gamma production from developing TH1 cells and is preferentially expressed by TH2 cells. Furthermore IL-21 has been identified as a growth and survival factor for human myeloma cells. IL-21/IL-21R interactions have a unique role in sequentially activating both innate and adaptive immune responses against poorly immunogenic tumors, leading to tumor rejection that is perforin dependent but IFN-gamma independent.

Human Recombinant IL15 (from CHO cells)
Supplier: Adipogen
Interleukin-15 (IL-15) has a broad spectrum of biological activities. It is crucial for the development, proliferation, survival and differentiation of multiple cells from both innate and adaptive immune systems. IL-15 up-regulation has a central role in the development of several autoimmune or chronic inflammatory disorders. Targeting IL-15 or its receptor may have a valuable impact on the treatment of immune-mediated diseases. IL-15 participates in the development of important immune antitumor mechanisms. It activates CD8(+) T cells, natural killer (NK) cells, NK T cells, and can promote the formation of antitumor antibodies. IL-15 can also protect T effector cells from the action of T regulatory cells and reverse tolerance to tumor-associated antigens. In pre-clinical studies IL-15 has been found to demonstrate potentiated antitumor effects following pre-association with IL-15Ralpha, or when used in combination with chemotherapy, adoptive therapy, monoclonal antibodies, and tumor vaccines.

Human Recombinant FGF-1 (from E. coli)
Supplier: Adipogen
Fibroblast growth factors (FGFs) constitute a family of heparin-binding polypeptides involved in the regulation of biological responses such as growth, differentiation and angiogenesis. The biological effects of FGFs are mediated by four structurally related receptor tyrosine kinases denoted FGFR1, FGFR2, FGFR3 and FGFR4. FGF-1 is a powerful mitogen of cells of mesodermal, ectodermal and endodermal origin. FGF-1 association with heparan sulfate is a prerequisite for activation of FGF receptors. FGF-1 plays a role in various stages of development and morphogenesis as well as in angiogenesis and wound healing processes. Recent data indicate a role of FGF-1 in inflammation and obesity. FGF-1 is selectively induced in fat cells by high-fat diet feeding and established the PPARgamma-FGF-1 axis as a critical pathway that regulates adipose tissue remodeling.

Anti-NW Monoclonal Antibody [clone: 117C]
Supplier: Adipogen
Reactive nitrogen species (RNS) are formed in a variety of diseases such as atherosclerosis, inflammation, cancer and neurodegenerative diseases. Peroxinitrite (ONOO-) is known as a potent biological oxidant, which is formed via very fast reaction between nitric oxide (NO) and superoxide (O2-). Peroxinitrite can induce nitration of aromatic amino acids such as Tyr, Trp, Phe and His, which results in modulation of protein functions. To date, 6-nitrotryptophan (6-NO2-Trp) is shown to be one of the most stable and major peroxynitrite-modified amino acids, and growing evidences suggest that nitrotryptophan may play a significant role in the cellular regulation under nitrosative stress. This antibody is suitable for immunohistochemistry, western blotting and ELISA and may provide a powerful tool for the investigation of nitrosative stress inside living bodies.

Anti-MLST8 Rabbit Polyclonal Antibody
Supplier: Rockland Immunochemical
GbetaL (G protein beta protein subunit-like) is a member of a signaling pathway that regulates mammalian cell growth in response to the presence of nutrients and growth factors. It binds to the kinase domain of TOR (Target of rapamycin, also known as mTOR), an evolutionarily conserved serine/threonine kinase that regulates cell growth and cell cycle through its ability to integrate signals from nutrient levels and growth factors. Rapamycin inhibits TOR resulting in reduced cell growth and reduced rates of cell cycle and cell proliferation. TOR is normally associated with GbetaL and an additional regulatory protein RAPTOR, allowing TOR to control protein biosynthesis. The binding of GbetaL to TOR stimulates TOR's kinase activity towards downstream proteins such as RPS6K (ribosomal protein S6 kinase) and the translation factor 4E-BP1 which leads to increased protein translation and cell growth.
