DARC, also known as the Duffy antigen/chemokine receptor, is a seven-transmembrane protein homologous to the classical chemokine G-protein coupled receptors (GPCRs) with the exception of the motif required for G protein coupling. DARC can bind with high affinity several chemokines without transducing any signal, suggesting it may modulate the signals normally induced by these chemokines. Recently, DARC was found to interact with KAI1, a four transmembrane protein recently identified as a tumor metastasis suppressor protein. It is thought that tumor cells dislodged from the primary tumor and expressing KAI1 interact with DARC proteins expressed on vascular cells, transmitting a senescent signal to the tumor cells, while tumor cells that have lost KAI1 expression can proliferate and potentially give rise to metastases. At least three isoforms of DARC are known to exist.

Interleukin-35 (IL-35) is a novel IL-12 family cytokine produced by regulatory T cells (Treg) but not by resting or activated effector T cells (Teff). IL-35 is a heterodimeric protein composed of EBI3 (Epstein-Barr-Virus-induced gene 3) and IL-12a (p35). EBI3 is a downstream target of Foxp3, a transcription factor required for Treg-cell development and function, and thus Treg-cell restriction of IL35 occurs. Regulatory T cells are essential for maintaining self tolerance and preventing autoimmunity, and IL-35 is identified as a molecule that mediates the immune suppression function of Treg-cell. As an inhibitory cytokine, IL-35 induces proliferation of Treg-cell populations but suppresses Th17 cell development. Studies in mice show the absence of either IL-35 chain from Treg-cell reduces the cells' ability to suppress inflammation using an experimental model for inflammatory bowel disease. IL-35 is suggested as a potential target of immunotherapy.

HES1 is well known in development of different tissues, particularly endoderm derived endocrine organs like pancreas and neuroendocrine cells of lung. HES1 controls normal development by regulating proliferation and differentiation and so cell fate. Notch signaling plays both oncogenic and tumor suppressor roles depending on cell type. In contrast to T cell acute lymphoblastic leukemia (ALL), where Notch activation promotes leukemogenesis, induction of Notch signaling in B cell ALL (B-ALL) leads to growth arrest and apoptosis. The Notch target HES1 is sufficient to reproduce this tumor suppressor phenotype in B-ALL by interacting and activating PARP-1 and regulating proapoptotic signals.

CD279 (Programmed Cell Death Protein 1; PD-1) is a type I transmembrane protein belonging to the CD28/CTLA-4 family of immunoreceptors that mediate signals for regulating immune responses. Members of the CD28/CTLA-4 family have been shown to either promote T cell activation (CD28 and ICOS) or downregulate T cell activation (CTLA-4 and PD-1). CD279 is expressed on activated T cells, B cells, myeloid cells and on a subset of thymocytes. In vitro, ligation of CD279 inhibits TCR-mediated T cell proliferation and production of IL-1, IL-4, IL-10 and IFN-gamma. In addition, CD279 ligation also inhibits BCR mediated signaling. Furthermore, down-regulation of Ambra1 in cultured cells though RNA interference decreased the level of rapamycin- and nutrient starvation-induced autophagy. Multiple isoforms of Ambra1 are known to exist.

Multiple isoforms of ATG16 are known to exist.
