CTHRC1 is expressed in calcified atherosclerotic plaque and chondrocyte-like cells, in the hypothalamus, pituitary gland and remodeling tissues. Aberrant expression of CTHRC1 is present in human solid cancers and likely to be associated with cancer invasion and metastasis. With adiponectin and other members of the C1q/TNF-related protein (CTRP) family, CTHRC1 shares the presence of a short N-terminal collagen domain but it lacks the characteristic C1q domain that defines the CTRP family of proteins. A recent study identifies CTHRC1 as a novel circulating hormone with metabolic effects.

In Drosophila cells, inactivation of CDCA8 results in polyploidy, delayed mitosis and abnormal tissue development, indicating its critical role for cell proliferation. Recent studies show that CDCA8 is essential for cell proliferation during early embryonic development, and its early embryonic lethality cannot be rescued by the loss of p53. Its aberrant expression is linked to a poor prognosis for gastric cancer.

High levels of CCR3 and CXCR4 expression were found on the neurons from both the central and peripheral nervous systems. The CCR3 ligand, eotaxin, and an anti-CCR3 antibody inhibited HIV infection of microglia. These results indicate CCR3 plays an important role in HIV infection of CNS.

The CDKN2A locus gives rise to 2 distinct transcripts from different promoters. The transcripts have been designated p16(INK4A) and p14(ARF). Conversely, transfection of CDKN2A suppressed the growth of two independent mesothelioma cell lines, suggesting that inactivation of the CDKN2 gene is an essential step in the etiology of malignant mesotheliomas. CDKN2A induces a G1 cell cycle arrest by inhibiting the phosphorylation of the Rb protein by the cyclin-dependent kinases CDK4 and CDK6. CDKN2A is expressed as at least three distinct isoforms.

These death signals finally cause the degradation of chromosomal DNA by activated DNase. A mouse DNase that causes DNA fragmentation was identified recently and designated CAD for caspase activated deoxyribonuclease. The human homologue of mouse CAD was more recently identified by three groups independently and termed CPAN, DFF40, and human CAD, respectively. DFF45/ICAD is the inhibitory protein of DFF40/CAD and forms complex with DFF40/CAD. Upon cleavage of DFF45/ICAD by activated caspase, DFF40/CAD is released and activated and eventually causes the degradation of DNA in the nuclei. Activation of DFF40/CAD, which causes DNA degradation, is the hallmark of apoptotic cell death.

More recently, DISC1 has been found to regulate the transport of a complex containing Nudel, the lissencephaly-1 (LIS1) protein, and 14-3-3epsilon from neuronal cell bodies to the axons by the action of the microtubule-dependent directed motor protein kinesin-1, also known as KIF5A. Decreased expression of DISC1 in neurons caused an accelerated rate of neuronal integration, resulting in aberrant morphological development, suggesting that DISC1 plays a role in dendritic development and synapse formation. DISC1 has at least four known isoforms.

A novel molecule that associates with and activates IKK was recently identified and designated CIKS (for connection to IKK and SAPK/JNK) and Act1 (for NF-kB activator 1). CIKS directly interacts with IKKgamma. CIKS/Act1 also activates activating transcription factor (ATF) and activator protein 1 (AP-1) through Jun kinase (JNK). These results indicate that CIKS/Act1 is involved in the inflammation and stress responses. CIKS/Act1 is ubiquitously expressed in human tissues.
