FOXO4 is a ubiquitously expressed protein member of a subfamily of the forkhead homeotic gene family of transcription factors and shuttles between the cytoplasm and nucleus. FOXO transcription factors are key players of cell fate decisions, metabolism, stress resistance, tumor suppression and are regulated by growth factors, oxidative stress or nutrient deprivation. In the absence of PI3K/AKT activation, FOXO4 localizes in the nucleus where it functions as a transcription factor. FOXO4 can also be phosphorylated by JNK following induction of reactive oxygen species (ROS), resulting in transcriptional activation and the induction of a negative feedback mechanism to counteract the ROS. It is through this mechanism that FOXO4 is thought to sensitize cancer cells to doxorubicin-mediated toxicity.

Fasciculation and elongation protein zeta-2 (FEZ2) is a homolog to the mammalian FEZ1, itself an ortholog of the C. elegans UNC-76. In contrast to FEZ1, FEZ2 mRNA is widely expressed in mouse tissues. FEZ2 interacts with protein kinase C (PKC)-zeta substrate and induces neurite extension of PC12 cells when co-expressed with a constitutively active form of PKC-zeta, suggesting FEZ2 may play an important role in the morphological changes of various cells by associating with PKC-zeta in a tissue non-specific manner. EFHD2, also known as Swiprosin-1 or SWS1, is an EF-hand and coiled-coil-containing adaptor protein that plays a role in lymphocyte physiology. EFHD2 exhibits the highest expression in CD8+ T cells and immature B cells. It provides a membrane scaffold that is required for the Syk-, SLP-65-, and PLCgamma2-dependent B-cell receptor (BCR)-induced calcium flux. FGFR3 is a tyrosine-protein kinase that acts as cell-surface receptor for fibroblast growth factors and plays an essential role in the regulation of cell proliferation, differentiation and apoptosis (1). It is required for normal skeleton development and promotes apoptosis in chondrocytes and cancer cell proliferation. FGFR3 protein contains three immunoglobulin-like domains, a single hydrophobic membrane-spanning segment and a cytoplasmic tyrosine kinase domain (1,2). It is alternatively spliced to produce three isoforms that are expressed in brain, kidney and testis. Defects in FGFR3 are associated with several diseases, including achondroplasia and hypochondroplasia (3,4). Mutations in FGFR3 are also a cause of some bladder and cervical cancers (5).

Apoptosis is induced by certain cytokines including TNF and Fas ligand of the TNF family through their death domain containing receptors, TNFR1 and Fas. A novel cell death receptor was recently identified by several groups independently and designated DR3, Wsl-1, Apo-3, TRAMP and LARD1-5. The ligand for this novel death receptor has been defined as TWEAK, also termed Apo3L. DR3 is highly expressed in the tissues enriched in lymphocytes including PBL, thymus and spleen. Like TNFR1, DR3 induces apoptosis and NF-kB activation.

Angiotensin-converting enzyme 2 (ACE2) plays a central role in vascular, renal, and myocardial physiology. In contrast to its homolog ACE, ACE2 expression is restricted to heart, kidney, and testis. Recently. ACE2 has also been shown to be a functional receptor of the SARS coronavirus. BHLHA15 (basic helix-loop-helix protein 15), also known as MIST1 (muscle intestine and stomach expression 1) belongs to the bHLH family of transcription factors and plays a role in regulating the transcriptional activity of MYOD1 in muscle cell development as well as serving as a key regulator of acinar cell function (1,2). BHLHA15 contains a basic helix-loop-helix (bHLH) domain and is capable of binding to E-box motifs as a homodimer or a heterodimer with E-proteins. It may also negatively regulate bHLH-mediated transcription through a N-terminal repressor domain (3). It is involved in the preservation of cellular nutrients under starvation conditions as well as the normal turnover of cytosolic components. This process is negatively regulated by TOR (Target of rapamycin) through phosphorylation of autophagy protein APG1. Another member of the autophagy family of proteins is ATG2B, one of two homologs of ATG2 that is essential for autophagosome formation and important for regulation of size and distribution of lipid droplets. Relatively high rates of ATG2B mutations were observed in gastric and colorectal carcinomas, suggesting that deregulating the autophagy process may contribute to cancer development.
