Aipl1 was initially identified as a protein implicated in Leber congenital amaurosis (LCA), an autosomal recessive disorder thought to be caused by the abnormal development of photoreceptors. Aipl1 is a tetratricopeptide repeat protein that is highly homologous to ARA9, a protein involved in the HSP90-mediated nuclear translocation and transactivation of the aryl hydrocarbon receptor. Aipl1 has also been found to function as part of a chaperone heterocomplex, interacting with Hsp90 and Hsp70. Aipl1 also associates with the cell cycle regulator NUB1. It is thought that Aipl1 cooperates with Hsp70 but not Hsp90 to suppress the formation of NUB1 inclusions, and these interactions are necessary in the normal photoreceptor maturation, as mutations that lead to LCA also compromise the interactions with the Hsp chaperones. At least three isoforms of Aipl1 are known to exist.

Collapsin-response mediator proteins (CRMPs) are highly expressed in the developing brain where they play major roles in axonal outgrowth, neurite differentiation, and apoptosis. Their continued expression in areas of high synaptic remodeling such as the cerebellum, hippocampus, and the olfactory system suggests that these proteins may also be involved in adult brain plasticity. CRMP-1 was initially identified as a dihydro-pyrimidinase expressed exclusively in brain; later studies have shown that it is involved with neurotrophin (NT) 3-induced neurite formation and outgrowth. CRMP-1 localization switches from axonal to somatodendritic when neurons reach functional maturity, suggesting that it is involved in early neuronal differentiation as well as in later processes related to the survival or death of the newly generated neurons.

Mitogen-activated protein (MAP) kinase cascades are activated in response to various extracellular stimuli, including cytokines, growth factors and environmental stresses. A novel MAP kinase kinase kinase (MAPKKK) was recently identified and designated ASK1 (for apoptosis signal-regulating kinase 1) and MAPKKK5. ASK1 activated two different subgroups of MAPKK, MKK4 and MKK6, which in turn activated c-Jun N-terminal kinase (JNK) and p38 MAP kinase, respectively. ASK1/MAPKKK5 is activated by TNFR and Fas through the interaction with members of the TRAF family and Fas-associated protein Daxx. Overexpression of ASK1 induced apoptotic cell death, and a catalytically inactive form of ASK1 inhibited TNF-a-induced apoptosis. ASK1 is expressed in variety of tissues and cell lines.

CCL22, also named stimulated T cell chemotactic protein (STCP1), is a CC chemokine initially isolated from clones of monocyte-derived macrophages and binds to the receptor CCR4. Human CCL22 is highly expressed in macrophage and in monocyte-derived dendritic cells and thymus. It is also found in lymph node, appendix, activated monocytes, resting and activated macrophages. May play a role in the trafficking of activated/effector T-lymphocytes to inflammatory sites and other aspects of activated T-lymphocyte physiology. CCL22 has been shown to induce chemotaxis or Ca2+ mobilization in dendritic cells, IL-2 activated NK cells and activated T lymphocytes.

Podoplanin is expressed on glomerular epithelial cells (podocytes), type I lung alveolar cells, lymphatic endothelial cells and numerous tumors, including colorectal tumors, squamous cell carcinomas, testicular seminoma and brain tumors. Podoplanin is the ligand for C-type lectin-like receptor 2 (CLEC-2). Their association is dependent on sialic acid on O-glycans of podoplanin. Through its association with CLEC-2, podoplanin induces platelet aggregation and tumor metastasis. Podoplanin is also necessary for lymphatic vessel formation, normal lung cell proliferation and alveolus formation at birth.

Progranulin (PGRN) is a widely expressed pluripotent growth factor which plays a role in processes such as development, wound repair and inflammation by activating signaling cascades that control cell cycle progression and cell motility. Its function in the central nervous system is of interest, as mutations in the PGRN gene were found in cases of frontotemporal degeneration (FTLD). Human CD28 is an important costimulatory molecule found on all CD4+ T cells and on about half of the CD8+ T cells. T cell activities attributed to CD28 include prevention of anergy, induction of cytokine gene transcription, stabilization of cytokine mRNA and activation of CD8+ cytotoxic T lymphocytes. The ligands for CD28 identified as CD80 (B7-1) and CD86 (B7-2) are immunglobulin superfamily monomeric transmembrane glycoproteins of 60kDa and 80kDa respectively. Anti-CD28 antibodies have been shown to have a strong adjuvant effect when conjugated to an immunogen. Monovalency of the CD28 receptor for its natural ligands is essential to provide costimulation without inducing responses in the absense of TCR engagement.

Interleukin-4 (IL-4) is a cytokine produced by type 2 helper T cells, the Th2 cells. These cells tends to make a specific set of lymphokines including IL-4, IL-5, IL-6, IL-10, IL-13, IL-3 and GM-CSF and fail to produce IL-2, IFN-gamma, and lymphotoxin (TNF-beta). In addition, mast cells can produce IL-4. IL-4 exerts numerous effects on various hematopoietic cell types. On B cells, IL-4 promotes immunoglobulin class switching to IgE and IgG1 isotypes and upregulates MHC class II and CD23 expression. IL-4 promotes survival, growth, and differentiation of both T and B lymphocytes, mast cells and endothelial cells. In addition, IL-4 inhibits the production of TNF, IL-1, and IL-6 by macrophages.

Human vascular endothelial erowth factor (VEGF 165) is produced as a homodimer. It binds to the FLT1/VEGFR1 and KDR/VEGFR2 receptors, heparan sulfate and heparin. VEGF-A is a specific mitogen for vascular endothelial cells and a strong angiogenic factor in vivo. In addition to its action as a mitogen it is a potent vascular permeability factor (VPF) in vivo. Nod (CARD4) is a cytosolic pattern recognition molecule (PRRs) of the NLR (NOD-like receptor) family.
