Apoptosis plays a major role in normal organism development, tissue homeostasis, and removal of damaged cells. Disruption of this process has been implicated in a variety of diseases such as cancer. Aven is a recently discovered protein that blocks apoptosis induced by Apaf-1 and caspase-9. It is thought that Aven functions by binding to Bcl-xL, an antiapoptotic member of the Bcl-2 family, and to Apaf-1, possibly interfering with the ability of Apaf-1 to self-associate, suggesting that Aven impedes Apaf-1-mediated caspase activation. Higher levels of Aven mRNA are seen in patients with acute leukemia than in control patients, suggesting that Aven may be useful as a prognostic indicator in leukemia patients.

CHD proteins belong to a superfamily of proteins of ATP-dependent chromatin remodeling enzymes that have a unique combination of functional domains, including two N-terminal chromodomains, a SNF2-like ATPase/helicase domain and a DNA-binding domain. These proteins are thought to play a role in early embryonic development by affecting chromatin structure and gene expression. Mutations in one member of this family, CHD7, result in CHARGE syndrome. It colocalizes with embryonic stem (ES) cell master regulators OCT4/POU5F1, SOX2, and NANOG and is thought to modulate ES-specific gene transcription. Together with SOX2, CHD7 has been suggested to also regulate several human disease genes.

Clathrin is a major protein component of the polyhedral coat of coated pits and vesicles. Two different adapter protein complexes link the clathrin lattice to either the plasma membrane or the trans-Golgi network. These specialised organelles are involved in the intracellular trafficking of receptors and endocytosis of a variety of macromolecules. Clathrin triskelions, composed of 3 heavy chains and 3 light chains, are the basic subunits of the clathrin coat. In the presence of light chains, hub assembly is influenced by both the pH and the concentration of calcium. Clathrin localises to the cytoplasmic face of coated pit and vesicle membranes.

CCAAT enhancer binding proteins (CEBPs) are a family of transcription factors that all contain a highly conserved, basic-leucine zipper domain at the C-terminus that is involved in dimerization and DNA binding. At least six members of the family have been isolated and characterized to date (CEBP alpha to CEBP zeta). CEBPD is a leucine zipper (LZ) DNA-binding protein that regulates gene expression in a variety of tissues including liver, adipose, lung and intestine. CEBPD is an important transcriptional activator in the regulation of genes involved in immune and inflammatory responses and has been reported to possess many tumor suppressor-like properties.

BRCC45 was initially suggested to be a housekeeping protein that is highly expressed in brain and reproductive organs. Later experiments indicated BRCC45 forms a complex with the breast and ovarian predisposition proteins BRCA1 and BRCA2 as well as RAD51 and BRCC36. This complex has a ubiquitin E3 ligase activity and is thought to enhance cellular survival following DNA damage. BRCC45 has also been suggested to function as a death receptor-associated anti-apoptotic protein by inhibiting the BID-induced activation of the mitochondrial apoptotic pathway. Higher levels of BRCC45 were detected in the majority of hepatocellular carcinomas, suggesting that BRCC45 may promote tumorigenesis when overexpressed. At least three isoforms of BRCC45 are known to exist.

BRAL1 is a member a superfamily consisting of several highly homologous hyaluronan and proteoglycan binding link proteins. BRAL1 is predominantly expressed in brain tissue and spinal cord. Like other members in the link-module superfamily, BRAL1 contains an immunoglobulin-like fold and two proteoglycan tandem repeats (PTRs). Its mRNA expression pattern is similar to other lectican proteoglycans, suggesting that BRAL1 may act to stabilize the binding between the extracellular matrix molecule hyaluronan and brevican. Immunostaining of mouse brain showed BRAL1 expression at P20 in the white matter of the developing cerebellum and in myelinated fiber tracts in the adult brain, suggesting that expression starts when axonal myelination occurs.

The estrogen receptor-related factor (ERRF) is a recently identified protein whose expression is upregulated in multiple cell lines and primary breast cancer tumors. Increased ERRF expression was significantly associated with ER or progesterone receptor (PR) positivity even in HER2-negative tumors. This higher ERRF expression correlated with better disease-free survival and overall survival. In two ER-positive breast cancer cell lines, knockdown of ERRF suppressed cell growth in vitro and tumorigenesis in xenograft models, suggesting that ERRF plays a role in estrogen-ER-mediated breast cancer cell growth and may thus be a potential therapeutic target.

Chromatin condensation and nuclear fragmentation (CCNF) is the hallmark of apoptosis. CCNF is triggered by the activation of members of caspase family, caspase activated DNase (CAD/DFF40), and several novel proteins including AIF and CIDE. A new inducer of chromatin condensation was recently identified and designated Acinus (for apoptotic chromatin condensation inducer in the nucleus). Acinus is cleaved by caspase-3 and an additional unknown protease generating a small active peptide p17, which causes chromatin condensation in vitro when it is added to purified nuclei. Acinus also induces apoptotic chromatin condensation in cells. Acinus is ubiquitously expressed. Three different spliced forms of Acinus have been identified in human and mouse and designated AcinusL, AcinusS and AcinusS'.

BICD1 was initially identified as the human homolog to Drosophila Bicaudal-D protein that forms part of the cytoskeleton and mediates the correct sorting of mRNAs for oocyte- and axis-determining factors during oogenesis. BICD1 can bind to dynein-dynactin and co-localizes with Rab6A on the trans-Golgi network and cytoplasmic vesicles, suggesting BICD1 regulates COPI-independent Golgi-ER transport. BICD1 will also interact with Rab6B, a Rab6 isoform that is expressed primarily in brain. Live cell imaging showed bi-directional movement of Rab6B structures in SK-N-SH neurites, indicating that BICD1 is involved in the Rab6B regulation of retrograde cargo transport in neuronal cells.
