Anti-Netrin-4 Mouse Monoclonal Antibody [clone: Nely-1]
Supplier: Adipogen
Netrin-4 is a 628 amino acid laminin-related secreted protein that promotes neurite elongation at low concentrations but inhibits neurite extension at high concentrations. Recent evidence suggest that Netrin family members act as regulators of both neuronal and vascular growth and are implicated in controling epithelial cell branching morphogenesis in the pancreas, breast and lung. Netrin-4 inhibits proliferation of a variety of human tumor cells. Netrin-4 also induces proliferation, migration, adhesion, tube formation and survival of human lymphatic endothelial cells and seems to function as a lymphangiogenic factor contributing to tumor dissemination.

Human Recombinant PARP-3 (from Insect Cells (Sf21))
Supplier: Adipogen
PARP-3 is involved in the base excision repair (BER) pathway, by catalyzing the poly(ADP-ribosyl)ation of a limited number of acceptor proteins involved in chromatin architecture and in DNA metabolism. This modification follows DNA damages and appears as an obligatory step in a detection/signaling pathway leading to the reparation of DNA strand breaks. May link the DNA damage surveillance network to the mitotic fidelity checkpoint. Negatively influences the G1/S cell cycle progression without interfering with centrosome duplication. May be involved in the regulation of PRC2 and PRC3 complex-dependent gene silencing.

Human Recombinant NMNAT2 (from E. coli)
Supplier: Adipogen
Nicotinamide mononucleotide adenylyltransferase 2 (NMNAT2) catalyzes the formation of NAD+ from nicotinamide mononucleotide (NMN) and ATP. Can also use the deamidated form nicotinic acid mononucleotide (NaMN) as a substrate but with lower efficiency. NMNAT2 also catalyzes the reverse reaction, i.e. the pyrophosphorolytic cleavage of NAD+. It is highly expressed in brain, in particular in cerebrum, cerebellum, occipital lobe, frontal lobe, temporal lobe and putamen. NMNAT2 is also found in the heart, skeletal muscle, pancreas and islets of Langerhans. NMNAT2 is essential for axon growth and survival. Its loss from injured axons may activate Wallerian degeneration (axon degeneration induced by nerve injury), whereas NMNAT overexpression rescues axons from degeneration.

Anti-DiBrY Monoclonal Antibody [clone: 3A5]
Supplier: Adipogen
Neutrophils and eosinophils play an important role in the defence system against microbial infection. Myeloperoxidase (MPO) and eosinophil peroxidase (EPO) are known to catalyze formation of hypochlorous acid (HOCl) and hypobromous acid (HOBr). These reactive intermediates may react with proteins, lipids and nucleotides, and it has been reported to form tyrosine halogenation such as dibromotyrosine (DiBrY). DiBrY is a Br-modified tyrosine at 3- and 5- position, which is one of the major oxidative products derived from neutrophil myeloperoxidase. DiBrY may be a novel biomarker for tissue damage which is related to inflammatory and allergic disorders.

Mouse Recombinant PEDF (from CHO Cells)
Supplier: Adipogen
Pigment epithelium-derived factor (PEDF) is a 47kDa secreted glycoprotein that belongs to the non-inhibitory serpin family group. PEDF is widely expressed in adult and fetal tissues, including brain, spinal cord, plasma, bone, prostate, pancreas, heart and lung. PEDF acts as an angiogenesis inhibitor with neurotrophic, immunomodulation and antitumor properties. It functions as anti-angiogenic agent by counterbalancing the proangiogenic effect of VEGF. PEDF is one of the most abundant proteins released by adipocytes and induces insulin resistance in adipocytes and human skeletal muscle cells. Recently, it has been reported that PEDF is sufficient to maintain the self-renewal of pluripotent human embryonic stem cells.

Human Recombinant POP (from E. coli)
Supplier: Adipogen
The inflammasome is a multiprotein complex that mediates the activation of caspase-1, which promotes, amongst others, the secretion of the proinflammatory cytokines interleukin (IL)-1beta and IL-18. Members of the Nod-like receptor family (NLRs), including NLRP3/NALP3, are critical components of the inflammasome that link danger-signals to caspase-1 activation. Defects in NLRP3 are the cause of familial cold autoinflammatory syndrome type 1 (FCAS1), Muckle-Wells syndrome (MWS) and of chronic infantile neurologic cutaneous and articular syndrome (CINCA). Pyrin domain-containing protein 1 (POP1) interacts with the protein Asc (Pycard) and modulates the interaction of Asc to NLR proteins.

Human Recombinant IL33 (from E. coli)
Supplier: Adipogen
Interleukin-33 (IL-33; HF-NEV; IL-1F11), a member of the IL-1 family of cytokines, is expressed by many cell types following pro-inflammatory stimulation and is thought to be released on cell lysis. The 30kDa human IL33 is converted by CASP1 to a 18kDa protein. IL33 binds to and signals through ST2 (IL1R1) and its stimulation recruits MYD88, IRAK, IRAK4, and TRAF6, followed by phosphorylation of ERK1 (MAPK3)/ERK2 (MAPK1), p38 (MAPK14), and JNK. The ability of IL-33 to target numerous immune cell types, like Th2-like cells, mast cells, and B1 cells, and to induce cytokine and chemokine production underlines its potential in influencing the outcome of a wide range of diseases, such as arthritis, asthma, atopic allergy & anaphylaxis, cardiovascular disease/atherosclerosis, nervous system diseases, and sepsis.

Anti-FLIP Rat Monoclonal Antibody [clone: Dave-2]
Supplier: Adipogen
FLIP is an apoptosis regulator protein which functions as a crucial link between cell survival and cell death pathways in mammalian cells and acts as an inhibitor of TNFRSF6 mediated apoptosis. A proteolytic fragment (p43) is likely retained in the death-inducing signaling complex (DISC) thereby blocking further recruitment and processing of caspase-8 at the complex. Full length and shorter isoforms have been shown either to induce apoptosis or to reduce TNFRSF-triggered apoptosis. FLIP lacks enzymatic (caspase) activity. FLIP is highly expressed in skeletal muscle, pancreas, heart, kidney, placenta and peripheral blood leukocytes.

Human Recombinant TRAIL (soluble) (from E. coli)
Supplier: Adipogen
TNF-related apoptosis-inducing ligand (TRAIL; Apo2L;CD253; TNFSF10) is a type II transmembrane protein of about 34kDa. Like most members of the tumor necrosis factor (TNF) superfamily of cytokines TRAIL can be cleaved at the cell surface by metalloproteases to form a soluble molecule. Active TRAIL forms trimers and specifically binds to fi ve distinct receptors: TRAIL-R1 (DR4; Apo2;CD261; TNFRSF10A), TRAIL-R2 (DR5; KILLER; TRICK2A;TRICK2B; CD262; TNFRSF10B), TRAIL-R3 (DcR1;LIT; TRID; CD263; TNFRSF10C), TRAIL-R4 (DcR2;TRUNDD; CD264; TNFRSF10D), and osteoprotegerin (OPG; OCIF; TNFRSF11B). Trimerized TRAIL triggers apoptosis upon ligation of cell surface TRAIL-R1 and/or TRAIL-R2 by inducingthe formation of the so-called multiprotein death-inducing signaling complex (DISC).

Anti-CD28 Monoclonal Antibody (FITC) [clone: ANC28.1/5D10]
Supplier: Adipogen
Human CD28 is an important costimulatory molecule found on all CD4+ T cells and on about half of the CD8+ T cells. T cell activities attributed to CD28 include prevention of anergy, induction of cytokine gene transcription, stabilization of cytokine mRNA and activation of CD8+ cytotoxic T lymphocytes. The ligands for CD28 identified as CD80 (B7-1) and CD86 (B7-2) are immunglobulin superfamily monomeric transmembrane glycoproteins of 60kDa and 80kDa respectively. Anti-CD28 antibodies have been shown to have a strong adjuvant effect when conjugated to an immunogen. Monovalency of the CD28 receptor for its natural ligands is essential to provide costimulation without inducing responses in the absense of TCR engagement.

Human Recombinant PARP-1 (from Insect Cells (Sf21))
Supplier: Adipogen
PARP-1 (ARTD1) is involved in the base excision repair (BER) pathway, by catalyzing the poly(ADP-ribosyl)ation of a limited number of acceptor proteins involved in chromatin architecture and in DNA metabolism. This modification follows DNA damages and appears as an obligatory step in a detection/signaling pathway leading to the reparation of DNA strand breaks. PARP-1 positively regulates the transcription of MTUS1 and negatively regulates the transcription of MTUS2/TIP150. It forms a complex with EEF1A1 and TXK that acts as a T-helper 1 (Th1) cell-specific transcription factor and binds the promoter of IFN-gamma to directly regulate its transcription, and is thus involved importantly in Th1 cytokine production.

Anti-CD40 Monoclonal Antibody (FITC) [clone: EA-5]
Supplier: Adipogen
CD40 belongs to the TNF-receptor superfamily and is essential in mediating a broad variety of immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development and germinal center formation. The interaction of CD40-CD40L is necessary for amyloid-beta-induced microglial activation, and thus is thought to be an early event in Alzheimer disease pathogenesis. CD40 is constitutively expressed by antigen presenting cells, including dendritic cells, B cells and macrophages. Consistent with its widespread expression on normal cells, CD40 is also expressed on a wide range of tumor cells, including non-Hodgkin's and Hodgkin's lymphomas, myeloma and some carcinomas including nasopharynx, bladder, cervix, kidney and ovary.

Human Recombinant BMP2 (from CHO cells)
Supplier: Adipogen
Bone morphogenetic protein 2 (BMP-2) is a member of the BMP subgroup of the TGF-beta superfamily. It plays a dominant role in embryonic dorsalventral patterning, organogenesis, limb bud formation and bone formation and regeneration. BMP-2 signals through heterodimeric complexes composed of a type I receptor (Activin RI, BMP-RIA or BMP-RIB) and a type II receptor (BMP-RII or Activin RIIB). BMP-2 induces chondrocyte proliferation, endochondral bone formation, longitudinal bone growth and bone and cartilage repair. It induces ectopic bone formation or calcification by promoting osteogenic and chondrogenic differentiation in mesenchymal cells, stem cells and vascular smooth muscle cells. It also promotes the maintenance and repair of colonic epithelium, suppresses neuronal dopamine synthesis and release, induces apoptosis in medulloblastoma cells and is required for cardiac contractility.
