Androgen-induced 1 (AIG1) is a novel Pirh2-interacting protein, which was initially cloned from human dermal papilla cells and is evolutionally conserved from drosophila through to humans. It is a membrane protein and belongs to the protein family containing FAR-17a domain. AIG1 may play a role in androgen-regulated growth of hair follicles. AIG1 could serve as a new biomarker for the diagnosis and prognostic evaluation of HCCs and may be a novel candidate tumor suppressor.

CCL17, also known as TARC, binds to chemokine receptors CCR4 and CCR8 and plays important roles in T cell development in thymus as well as in trafficking and activation of mature T cells. CCL17 is secreted from monocyte-derived DCs and endothelial cells and is responsible for selective recruitment and migration of activated Th2 lymphocytes. The CCR7 protein is a member of the G protein-coupled receptor family. This receptor was identified as a gene induced by the Epstein-Barr virus (EBV), and is thought to be a mediator of EBV effects on B lymphocytes. DISP1 is the mammalian homolog of the Drosophila Dispatched segment-polarity gene and a key regulator of the Hedgehog (Hh) signaling pathway during embryonic development. DISP1 is required to move Shh from site of synthesis during embryogenesis; mutations in the gene result in lethality at mid-gestation and prevents specification of ventral cell types in the neural tube. DELE is a recently identified DAP3-binding protein that is thought to be important in the induction of death receptor-mediated apoptosis. Transfected cells that stably express DELE were found to be susceptible to apoptosis induction by TNF-a and TRAIL, whereas reducing DELE expression by siRNA rescued these cells from apoptosis induction. Furthermore, the reduction of DELE expression also inhibited the activation of caspase-3, caspase-8 and caspase-9 following stimulation by TNF-a, anti-Fas, or TRAIL, indicating the importance of DELE in apoptosis mediated by death receptors.

Ethidium Homodimer, first developed by Dr. Le Pecq and his colleagues, is a high-affinity fluorescent nucleic acid stain. It binds to both DNA and RNA in a sequence-independent manner and with a >30-fold fluorescence enhancement. The DNA binding of each Ethidium Homodimer covers four base pairs and is believed to occur by intercalation. Because the dye is highly positively charged, it cannot cross cell membranes to stain living cells. It is useful for detecting nucleic acids in solution, or for selectively staining dead cells with damaged plasma membranes.

Although FITC reagents have been more often used to prepare fluoresceinated bioconjugates, the low stability of FITC bioconjugates makes some researchers use amine-reactive succinimidyl esters of carboxyfluorescein (commonly called FAM) in bioconjugations. FAM reagents give carboxamides that are more resistant to hydrolysis. We have shown that FAM reagents require less stringent reaction conditions and give better conjugation yields, and the resulted conjugates have superior stability.
