Interleukin-2 (IL-2) is a 133 amino acid glycoprotein with one intramolecular disulfide bond and variable glycosylation. It is secreted by activated T cells and induces proliferation and maturation of activated T cells, natural killer cells, and lymphokine activated killer cells. IL-2 also stimulates proliferation of antibody-producing B cells, activates neutrophils, and induces mononuclear cells to secrete IFN-gamma and TNF-alpha and -beta. Moreover, studies have shown that IL-2 is required for activation-induced apoptosis, an important hemeostatic mechanism in the immune system, which is involved in the maintenance of peripheral tolerance to self-antigens.

Human CD4 is a cell surface glycoprotein of 55kDa expressed on most thymocytes, on about two thirds of peripheral T cells and on some monocyte macrophage lineage cells. CD4 interacts with MHC Class II molecules in an accessory role during foreign antigen recognition by T cells. The extracellular portion of CD4 is an array of four Ig-like domains. The N-terminal region of CD4 is a receptor for the HIV-1 viral protein gp120.

MHC Class I molecules consist of heterodimers of a highly polymorphic transmembrane alpha chain and an invariant beta chain. Trypan Blue is a blue acid dye with a strong affinity for cellulose containing substrates such as cotton; less affinity for proteinaceous materials. Trypan blue solution may be used in trypan blue based cytotoxicity and proliferation assays.
Trypan Blue is used as a vital dye which is especially important because it is taken up by the reticuloendothelial system. Clark describes assays for the study of teratogenic action of trypan blue on embryonic tissues using Davis and Sauter's fluorescence method and for the staining of collagen, including very fine fibrils, muscle and cornified epithelium using the Van Gieson method. Trypan blue is also recommended for use in dye exclusion procedures for viable cell counting. Non-viable cells will up-take trypan blue at a faster rate than viable cells.

Ethidium Homodimer, first developed by Dr. Le Pecq and his colleagues, is a high-affinity fluorescent nucleic acid stain. It binds to both DNA and RNA in a sequence-independent manner and with a >30-fold fluorescence enhancement. The DNA binding of each Ethidium Homodimer covers four base pairs and is believed to occur by intercalation. Because the dye is highly positively charged, it cannot cross cell membranes to stain living cells. It is useful for detecting nucleic acids in solution, or for selectively staining dead cells with damaged plasma membranes.

The CCR7 protein is a member of the G protein-coupled receptor family. This receptor was identified as a gene induced by the Epstein-Barr virus (EBV), and is thought to be a mediator of EBV effects on B lymphocytes. DELE is a recently identified DAP3-binding protein that is thought to be important in the induction of death receptor-mediated apoptosis. Transfected cells that stably express DELE were found to be susceptible to apoptosis induction by TNF-a and TRAIL, whereas reducing DELE expression by siRNA rescued these cells from apoptosis induction. Although FITC reagents have been more often used to prepare fluoresceinated bioconjugates, the low stability of FITC bioconjugates makes some researchers use amine-reactive succinimidyl esters of carboxyfluorescein (commonly called FAM) in bioconjugations. FAM reagents give carboxamides that are more resistant to hydrolysis. We have shown that FAM reagents require less stringent reaction conditions and give better conjugation yields, and the resulted conjugates have superior stability.
