Trichloroacetic acid is an acetic acid analogue commonly used to precipitate proteins, DNA and RNA. In the presence of SDS Trichloroacetic acid will precipitate proteins which can then be quantified by the Lowry method. The compound has also found uses as a decalcifier and fixative in microscopy, in protein sequencing, detecting albumin and organic synthesis.
Trichloroacetic acid is used in protein precipitation; has been used to determine protein concentration by quantitative precipitation. It is also used as a decalcifier and fixative in microscopy. A protocol for the precipitation of nucleic acids can be found in Molecular Cloning. Rats exposed to chronic sublethal amounts of trichloroacetic acid displayed an increase in serum bilirubin with a decrease in hematological proteins and cholesterols along with significant decreases in red blood corpuscles, mean cell volume, mean corpuscular hemoglobin, hemoglobin and hematocrit. In mice the production of liver tumors was associated to Trichloroacetic acid, which is also a known mouse hepatocarinogen. On the guinea pig trichloroacetic acid was observed to be a mild alergen on the skin.

The Pierce Silver Stain Kit is a rapid, ultra-sensitive, and versatile silver stain system for protein detection in polyacrylamide gels, yielding consistent and reliable results. It is a metallic silver (Ag) protein stain that yields a remarkably clear and uniform gel background. In standard mini gels, proteins are detectable at greater than 0.25 ng per band or spot. The protocol has been optimized for flexibility by allowing short or overnight gel fixation and staining steps without affecting staining performance (sensitivity or clarity). A short (one-minute) sensitization step, performed after gel fixation, yields results that are free of the characteristically dark or blotchy backgrounds often seen with homemade or other commercially available silver stains. This feature is beneficial when performing densitometric analysis of silver stained gels.

IL-6 is reported to be produced by fibroblasts, activated T cells, activated monocytes or macrophages and endothelial cells. It acts upon a variety of cells including fibroblasts, myeloid progenitor cells, T cells, B cells and hepatocytes. In addition, IL-6 appears to interact with IL2 in the proliferation of T lymphocytes. IL-6 potentiates the proliferative effect of IL-3 on multipotential hematopoietic progenitors. IL-6 is a multifunctional 24 kDa protein originally discovered in the medium of RNA stimulated fibroblastoid cells. It is upregulated by IL1, TNF, PDGF, IFN beta, TNF alpha, NGF, IL17 and downregulated by glucocorticoids IL4, TGF beta. IL-6 appears to be directly involved in the responses that occur after infection and cellular injury, and it may prove to be as important as IL1 and TNF alpha in regulating the acute phase response. IL6 has also been implicated in regulating adipose mass.

Ficin is a purified ficin preparation which is extracted from the latex of the fig tree Ficus glabrata. Ficin is classified as a thiol protease. Ficin hydrolyses the peptide bonds where the carbonyl group is from phenylalanine or tyrosine. When used in conjunction with other plants proteases, papain or bromelain, a synergistic effect may be observed. Immobilized Ficin was specifically designed for cleavage of mouse IgG1 into F(ab')2 or Fab fragments.The immobilization of ficin enhances stability against denaturation, heat and autolysis. IL4 is a pleiotropic cytokine produced by activated T cells, mast cells and basophils. It is a ligand for Interleukin 4 receptor. The Interleukin 4 receptor also binds to IL13, which may contribute to many overlapping functions of this cytokine and IL13. IL4 elicits many different biological responses, but has two dominant functions. The first is regulating differentiation of naïve CD4+ T cell to the Th2 type. Th2 cells produce IL4, IL5, IL10 and IL13, which tend to favor a humoral immune response while suppressing a cell mediated immune response controlled by Th1 cells. STAT6, a signal transducer and activator of transcription, has been shown to play a central role in mediating the immune regulatory signal of this cytokine. The second is regulating IgE and IgG1 production by B cells. Two alternatively spliced transcript variants of this gene encoding distinct isoforms have been reported.

Poly-lysine is a polycation which binds to DNA, red cell membrane and any negatively charged protein. When adsorbed to the culture surface, poly-lysine increases the number of positively charged sites available for cell binding. A compromise between the easier to use lower molecular weight products and the extremely viscous higher molecular weights would be the products in the range of 70,000-150,000.
It is typically used as a coating substrate for culture dishes, slides, etc. It enhances electrostatic interaction between negatively charged ions of the cell membrane and the culture surface. Both the D- and L- form of the poly-lysine can be used as a coating substrate since poly-lysine is a nonspecific attachment factor for cells; however, certain cells can digest poly-lysine. In this case, poly-D-lysine should be used as the attachment factor so that the cells are not disrupted by excessive uptake of L-lysine.
Other uses for poly-lysine have been reported as well:
• Conjugation to methotrexate for increased drug transport.
• Microencapsulation of islets.
• Use in simple reproducible procedure for chromosomal preparations from a variety of tissues.
• Immobilation of living cells in biocompatible semipermeable microcapsules.
• In the preparation of polycationic beads.
• Conjugation to albumin and horseradish peroxidase to enhance cellular uptake.

This colourless liquid is the simplest organic nitrile (hydrogen cyanide is a simpler nitrile, but the cyanide anion is not classed as organic). It is produced mainly as a byproduct of acrylonitrile manufacture.
Acetonitrile is widely used as a polar aprotic solvent in organic synthesis and in the purification of butadiene in refineries.
This solvent is often the preferred choice as a mobile phase in HPLC for a variety of reasons. Not only is it miscible with water and with many other organic solvents, but it also has a lower viscosity and produces less pressure in the column than other solvents such as Methanol. The inherent low absorbance of Acetonitrile (especially HPLC grade) makes it the preferred solvent for the mobile phase of HPLC due to the reduction in the amount of noise during UV detection. Overall, Acetonitrile has many unique properties as an organic mobile phase that often result in better chromatographic peaks.
Acetonitrile also plays an important role as the main solvent used in the manufacture of DNA oligonucleotides from monomers.

Iterleukin 7 is a lymphoid cell growth factor that affects pre-B, pro-B, and early T cells. IL-7 was previously known as pre-B cell growth factor and lymphopoietin 1. IL-7 supports the growth of early B cells from long-term lymphoid bone marrow cultures. It is mitogenic to thymocytes and enhances the response of cells to other stimuli such as polyhydroxyalkanoate (PHA) and concanavalin A (ConA). IL-7 stimulates the proliferation of CD4+/CD8+ cells. The proliferative response of thymocytes to IL-7 is not affected by antibodies to the T cell growth factors such as IL-2, IL-4 and IL-6, suggesting that IL-7 is capable of stimulating T cell proliferation through a pathway independent of the known T cell growth factors. Mature T cells respond to IL-7 and Con A, but not to IL-7 alone. In myeloid lineage cells, IL-7 upregulates the production of pro-inflammatory cytokines and stimulates the tumoricidal activity of monocytes/macrophages. IL-7 is expressed by adherent stromal cells from various tissues.
