US EPA SOW ILM 05.3/ISM 01.2 gives specific procedures for the methods of analysis, target elements, and concentrationlevels. Standards are specified not only by the elements present and their relative concentrations, but also the order and frequency of running standards, blanks and samples. CLP ISM 01.2 & ILM 05.3 Standards for ICP
For ILM 05.3, a standard must be run at the Contract Required Detection Limits (CRDL). To verify linearity near the CRQL, this standard is analyzed at the beginning of the analysis run, after the ICV/ICB and before the ICSA and ICSAB. In addition, this standard must be run at a frequency of not less than 20 analytical samples and at the end of the analysis run, followed by the ICSA/ISCAB. The sequence order is CCV, CCB, CRI, ICSA, ICSAB, CCV, and CCB. For ISM 01.2, at least one of your calibration standards must be at the Contract Required Detection Limit (CRDL). This standard, when diluted, can be used as a calibration standard to fulfill this requirement.

Initial Calibration Verification for CLP Methods
The US EPA retains analytical services through the Contract Laboratory Program (CLP). The CLP follows detailed SOPs derived from EPA methods. The CLP methods require calibration of analytical instrumentation whithin the expected quantitative range (ICAL standards) and additional CLP QA standards (ICV standards) to verify the calibration curve at each of the selected wavelengths that wil be used for sample analysis.

Our verification standards contain all of the elements on the TAL list and are independent standards for testing an instruments calibration curve. SPEX CertiPrep's ICV standards are designed to be used with their corresponding instrument calibration standards (ICAL).

We recommend dilution of ICV standards to a range within your instruments calibration curve. A dilution of 20-fold is recommended.

5-Bromo-4-chloro-3-indolyl β-D-galactopyranoside, commonly known as X−Gal, is a histochemical substrate for β−galactosidase.
5-Bromo-4-Chloro-3-Indolyl-β-D-Galactopyranoside is used as indigogenic substrate for β-galactosidase, for detection of β-galactosidase-positive clones, and the identification of lac and bacterial colonies or phage plaques. It is the substrate of choice for blue-white selection of recombinant bacterial colonies with the lac+ genotype. X−Gal is cleaved by β−galactosidase to yield an insoluble blue precipitate. X−Gal is particularly useful in molecular biology applications to detect the activity of β−galactosidase which is frequently used as a reporter gene. In cloning, X−Gal is used to detect insertion of foreign DNA into the lacZ region of plasmid DNA using α-complementation which is based on vectors such as the pUC and the M13mp series that carry a fragment of the β-galactosidase gene encoding an α-fragment of β-galactosidase.

Urea is the principal end product of nitrogen metabolism in most mammals, formed by the enzymatic reactions of the Kreb's cycle.
Urea is a mild agent usually used in the solubilization and denaturation of proteins. It is also useful for renaturing proteins from samples already denatured with 6 M guanidine hydrochloride such as inclusion bodies; and in the extraction of the mitochondrial complex. It is commonly used to solubilize and denature proteins for denaturing isoelectric focusing and two-dimensional electrophoresis and in acetic acid-urea PAGE gels. Urea is used in cell or tissue culture media to increase the osmolality. Urea has also been used as fertilizer because of the easy availability of nitrogen; in animal feeds; it is reacted with aldehydes to make resins and plastics; condensed with malonic ester to form barbituric acid; used in the paper industry to soften cellulose; used as a diuretic; enhances the action of sulfonamides; an antiseptic.
Urea in solution is in equilibrium with ammonium cyanate. The form that reacts with protein amino groups is isocyanic acid. Urea in the presence of heat and protein leads to carbamylation of the proteins. Carbamylation by isocyanic acid interferes with protein characterization because isocyanic acid reacts with the amino terminus of proteins, preventing N-terminal sequencing. Isocyanic acid also reacts with side chains of lysine and arginine residues resulting in a protein that is unsuitable for many enzymatic digests. In addition, carbamylation often leads to confusing results from peptides having unexpected retention times and masses. When performing enzymatic protein digests it is important to remove urea first. Even though some enzymes will tolerate small amounts of urea, the elevated temperature used for most reactions will lead to carbamylation during the course of the digest. The urea can be removed prior to digestion by fast reversed phase chromatography, spin columns, or dialysis.
