It is known that phenol and p-cresol are promoters of skin tumors and also urinary excretion of phenol and p-cresol is increased by exposure to benzene, which has hemopoietic toxicity, and toluene, respectively. Therefore, the determination of phenol and p-cresol is important in clinical chemistry and toxicology. Because phenol and p-cresol are present in urine as sulfate or glucuronide conjugates, they are hydrolyzed enzymatically or with acid, followed by distillation or extraction with organic solvent prior to measurement by GC or HPLC. However, these GC and HPLC methods require relatively large sample volumes (more than 1 ml) because of their insufficient sensitivities for measuring small amounts of phenol and p-cresol. In the general, the application of fluorescent labeling techniques to HPLC result in a sensitive and selective method. Some acyl chlorides are known to react with phenolic hydroxy group to form esters. It is known that 4-(N-phthalimidinyl)benzensulfonyl chloride Phisyl-Cl7, having sulfonyl chloride, reacted with phenol to give the corresponding highly fluorescent sulfonyl ester. The sensitivity of Physil-Cl was compared with Dansyl-Cl and DPS-Cl, which are analogous to Physil-Cl as fluorescent labeling reagents having sulfonyl chloride group. The labeling reaction of phenol with Dansyl-Cl and DPS-Cl were carried out according to the method of Ruiter et al.5 and the peaks were detected at the each fluorescence maxima wavelength (Ex350nm a Em550nm for Dansyl-Cl, and Ex320nm a Em386nm for DPS-Cl). Peak height of Phisyl derivative was higher about 85-and 7,3-fold than thosse of Dansyl derivative and DPS derivative, respectively. Among these three reagents, Phisyl-Cl was the most sensitive.

Anti-Human secondary antibodies are affinity-purified antibodies with well-characterized specificity for human immunoglobulins and are useful in the detection, sorting or purification of its specified target. Secondary antibodies offer increased versatility enabling users to use many detection systems (e.g. HRP, AP, fluorescence). They can also provide greater sensitivity through signal amplification as multiple secondary antibodies can bind to a single primary antibody. Most commonly, secondary antibodies are generated by immunizing the host animal with a pooled population of immunoglobulins from the target species and can be further purified and modified (i.e. immunoaffinity chromatography, antibody fragmentation, label conjugation, etc.) to generate highly specific reagents.

Peroxidase-conjugated goat IgG fraction to human IgG Fc is used in immunostaining of acetone-fixed frozen and formalin-fixed, paraffin-embedded tissue sections. It is also suitable for use as a reagent in enzyme immunoassays (EIA), cell and tissue staining (for light microscopy), cell and tissue labeling (for electron microscopy), and blot immunostaining. (Note: F(ab')2 fragments are recommended for staining of cells or tissues which contain Fc receptors. Affinity purified antibodies are recommended to avoid non-specific binding from inherent antibodies of host animals.

Antibody and highly purified HRP (Rz>3.0) are conjugated under defined conditions to obtain optimally labeled product. Conjugated protein is purified by salt fractionation. The product is dialyzed into 0.02M sodium phosphate, 0.14M sodium chloride, pH 7.3, with 0.01% thimerosal, adjusted to standard titer, filtered through a 0.22 μm filter, vialed and lyophilized.

The total protein is measured using the Biuret procedure with bovine albumin as the standard. To ensure that the product titer falls within the required range, antibody titer is standardized by microtiter plate ELISA with human IgG. The product is tested for purity and specificity at final concentration by immunoelectrophoresis.

This antibody recognizes a phosphor-protein of 45 kDa, identified as MyoD1. This MAb does not cross react with myogenin, Myf5, or Myf6. Antibody to MyoD1 labels the nuclei of myoblasts in developing muscle tissues. MyoD1 is not detected in normal adult tissue, but is highly expressed in the tumor cell nuclei of rhabdomyosarcomas. Occasionally nuclear expression of MyoD1 is seen in ectomesenchymoma and a subset of Wilm s tumors. Weak cytoplasmic staining is observed in several non-muscle tissues, including glandular epithelium and also in rhabdomyosarcomas, neuroblastomas, Ewing s sarcomas and alveolar soft part sarcomas.

Anti-KRT19 Mouse Monoclonal Antibody (CF405S) [clone: Ks19.1]

This MAb reacts with the rod domain of human cytokeratin-19 (CK19), a polypeptide of 40 kDa. Its epitope maps between amino acid 312-335. CK19 is expressed in sweat gland, mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, and ectocervical epithelium. Anti-CK19 reacts with a wide variety of epithelial malignancies including adenocarcinomas of the colon, stomach, pancreas, biliary tract, liver, and breast. Perhaps the most useful application is the identification of thyroid carcinoma of the papillary type, although 50%-60% of follicular carcinomas are also labeled. Anti-CK19 is a useful marker for detection of tumor cells in lymph nodes, peripheral blood, bone marrow and breast cancer.

CF® dyes are Biotium's next-generation fluorescent dyes. CF®405S is a blue fluorescent dye (Ex/Em 404/431 nm) with superior brightness compared to other blue dyes; it is also compatible with super-resolution imaging by SIM.
