PEDF is a potent inhibitor of angiogenesis. As it does not undergo the S (stressed) to R (relaxed) conformational transition characteristic of active serpins, it exhibits no serine protease inhibitory activity. The PEDF gene is a member of the serpin gene family. Serpins are a group of serine protease inhibitors, some of which have also been reported to exhibit neurotrophic activity.

Sodium chloride is a commonly used chemical which is found widely in nature. It is considered to be an essential nutrient. Excess amounts of sodium chloride can destroy electrolyte balance and cause death in most animals, including humans.
Sodium chloride is used in a wide variety of biochemical applications, including intravenous fluids (normal saline is 0.9% w/v in water 10), density gradients 3,6, a diluent to increase ionic strength in buffers or culture media and in salt-out procedures in the isolation of DNA. It has also been used in high concentrations for preservation of foods since bacteria cannot grow in high salt conditions. A salt-ice mixture in the ratio of approximately 33 g sodium chloride to 100 g ice (at -1°C) will drop in temperature to as low as -21°C, depending on the rate of stirring and the size of the ice chunks.

Deoxyribonuclease from beef pancreas, DNase I, was first crystallized by Kunitz. It is an endonuclease which splits phosphodiester linkages, preferentially adjacent to a pyrimidine nucleotide yielding 5'-phosphate terminated polynucleotides with a free hydroxyl group on position 3'. The average chain of limit digest is a tetranucleotide. DNase I acts upon single chain DNA, and upon double-stranded DNA and chromatin. In the latter case, although histones restrict susceptibility to nuclease action, over a period of time nearly all chromatin DNA is acted upon. According to Mirsky and Silverman, this could result from the looseness of histone attachment to DNA. They found that lysine-rich histones more effectively block DNase access to DNA than arginine-rich histones. Billing and Bonner suggest that DNase attacks the histone-free strand of chromatin DNA. Schmidt, et. al.indicate that hydrolysis of the histone-free region of DNA strands accounts for the initial rapid action of the enzyme on chromatin. Bollum reports degradation of synthetic homopolymer complexes by DNase I. The intracellular functions of the enzyme are probably controlled by a DNase inhibitor, which according to Lazarides and Lindberg is actin.

Guanidine Hydrochloride is a strong chaotropic agent useful for the denaturation and subsequent refolding of proteins. This strong denaturant can solubilize insoluble or denatured proteins such as inclusion bodies. This can be used as the first step in refolding proteins or enzymes into their active form. Urea and dithiothreitol (DTT) may also be necessary. Guanidine HCl is used in the isolation of RNA to dissociate the nucleoprotein into its nucleic acid and protein moieties. It is an inhibitor of RNase. Highly concentrated (6 - 8 M) Guanidine HCl solutions are used to denature native globular proteins. It apparently disrupts hydrogen bonds which hold the protein in its unique structure. However, there also is evidence suggesting that guanidine hydrocholoride may disrupt hydrophobic interactions by promoting the solubility of hydrophobic residues in aqueous solutions.

Tween® 80 has been widely used in biochemical applications including: solubilizing proteins, isolating nuclei from cells in culture selective protein extraction growing of tubercule bacilli, and emulsifying and dispersing substances in medicinal and food products. It has little or no activity as an anti-bacterial agent. It has been shown to have an adverse effect on the antibacterial effect of methyl paraben and related compounds. Non-ionic detergent used for selective protein extraction and isolation of nuclei from mammalian cell lines.
Soluble/miscible in water to give a clear yellow solution; miscible with alcohol, dioxane, and ethyl acetate; practically insoluble in liquid paraffin and fixed oils (such as mineral oil).
Autoclaving of solutions is not recommended. Sterile filtering is suggested with a 0.22 micron filter. Tween® may need to be warmed to about 40 °C and alternated with portions of hot distilled water while being poured through the filter.
Store at Room Temperature (15-30 °C).

The potent toxicity of both the botulinum neurotoxins and anthrax lethal toxin is due to a zinc-dependent proteolytic activity associated with the toxins. Measurement of this enzymatic activity provides for both a potentially sensitive and direct means for detection of the toxin, and a method for identifying potential toxin inhibitors using high throughput screening. A highly efficient approach for monitoring enzymatic activity is based on the use of fluorescence resonance energy transfer (FRET) substrates. These fluorogenic peptides contain a donor fluorescent group at one end and a suitable chromogenic acceptor group at the other. The fluorescence is quenched initially by intramolecular energy transfer between the donor/acceptor pair. Cleavage of the FRET substrate by the appropriate enzyme releases the fluorophore and full fluorescence is restored. The increase in fluorescence intensity is directly proportional to the amount of enzyme present. Enzymatic activity can be monitored continuously by recording the increase in fluorescence intensity with time. The change in the relative fluorescence units (RFU) as cleavage occurs can be converted to nmoles of cleaved substrate from a standard curve generated using a Calibration Peptide which is the cleaved substrate containing only the N-terminally attached fluorophore. For Botulinum neurotoxin type A, a Control FRET peptide substrate that is not cleaved by the neurotoxin but contains all remaining non-specific sites in the sequence can be used to screen background cleavage of the substrate that can occur in complex matrices.
