Interleukin 17A (IL-17A) is the founding member of the family of cytokines that includes IL-17B through IL-17F. It is a potent pro-inflammatory cytokine that plays a key role in defense against pathogens. IL-17A and IL-17F signal as homodimers or heterodimers through the same receptor, and activate NF-κB, MAPK, and C/EBP pathways (Gaffen). IL-17A is produced by Th17 cells, CD8+ T cells, γ/δ T cells, natural killer (NK) T cells, B cells, innate lymphoid cells, and mesenchymal stromal cells (MSCs) (Cua and Tato; Gaffen; Mojsilović et al.). IL-17A increases the frequency and the average size of fibroblast colony-forming units (CFU-F), as well as the proliferation of marrow-derived MSCs. Verapamil hydrochloride is an alpha-adrenergic receptor (alpha-AR) antagonist and calcium channel protein inhibitor that blocks the L-type Ca2+ channels in smooth and cardiac muscle cells. Verapamil is an antiarrhythmic agent and vasodilator known to reduce the renal clearance of digoxin and induce apoptosis in primary and metastatic colon adenocarcinoma human cell lines in vitro. Interleukin 6 receptor (IL-6R) alpha is a type I transmembrane glycoprotein that forms a complex with type I transmembrane signal transducer protein gp130 (CD130) and mediates the biological activities of IL-6. IL-6 binds to the membrane-bound non-signaling IL-6R alpha (mIL-6R), and the complex binds to two molecules of gp130 and leads to ‘classical’ IL-6-signal transduction, which includes activation of JAK/STAT, ERK, and PI3K signal transduction pathways (Scheller et al.). The 2,3-diaminonaphthalene (DAN) assay is routinely used in the determination of nitrite/nitrate levels in biological fluids and cellular extracts as one indicator of nitric oxide activity. The assay, as reported by Misko, uses the reaction of DAN with NO2- under acidic conditions to form a detectable fluorescent naphthotriazole. The reaction proceeds at acidic pH at room temperature. Fluorescence is monitored following the addition of NaOH, which raises pH, resulting in lower background and increased sensitivity (Ex/Em: ~365/415nm). However, detection at 450nm is recommended to avoid fluorescent blanks and increase sensitivity. The fluorescent background of DAN is low for maximum sensitivity. Detection limits of NO achieved by this method are in the high-pM range. 2,3-Diaminonaphtaline (DAN) is used as a derivatisation-reagent for the determination of selenium at low detection limits. MBQ-167 >98% Rac/Cdc42 inhibitor

Dual inhibitor of the Rho GTPases Rac (IC50=103 nM) and Cdc42 (IC50=78 nM).1 It inhibits breast cancer cell migration, viability, and mammosphere formation. MBQ-167 induced polarity loss resulting in 95% cell rounding and detachment from the substratum in metastatic MDA-MB-231 cells and was active in GFP-HER2-BM, MDA-MB-468, and Hs578t breast cancer cells as well as Mia-PaCa-2 pancreatic cancer cells, SKOV3 ovarian cancer cells, AGS and NCI-N87 gastric cancer cells, and SH-SY5Y neuroblastoma cells. Non-cancerous mammary epithelial MCF10A and epithelial breast cancer MCF-7 cells were resistant to MBQ-167. MBQ-167 inhibited viability and induced apoptosis in gefitinib and lapatinib resistant SKBR3 breast cancer cells.2 MBQ-167 inhibited triple negative breast cancer tumor growth and lung metastasis in a mouse model.3 Active in vivo.
