Neural Stem Cells (NSC) are self-renewing cells, of the neural lineage, capable of generating neurons (GABA, dopamine and motor neurons), astrocytes and oligodenrocytes. NSCs are marked by expression of Nestin and can be derived from fetal neural stem cells, adult neural stem cells, embryonic stem cells and adult somatic cells. NSCs are typically expanded and maintained in 2D monolayers or 3D neurosphere cultures containing recombinant EGF and FGF-basic (also called FGF-2 or FGFb). To differentiate NSCs to more specific cell types, the EGF and FGF-basic containing media is often replaced with media containing various cocktails of BDNF, SHH, FGF-8, CNTF, LIF, BMP-4, NT-3, NT-4, PDGFs and other cytokines. Recombinant human EGF and FGF-basic show cross-species reactivity so they are sometimes used in mouse and/or rat cultures.

Neurotrophin-3 (NT-3) is a neurotrophic factor and a member of the nerve growth factor (NGF) family of proteins that includes neuron growth factor (NGF), brain-derived neurotrophic factor (BDNF), and neurotrophin-4/5. NT-3 signals a number of trophic effects through its transducing receptor tyrosine kinase TrkC. NT-3 is known to promote survival, development, and differentiation of neurons, and modulates transmitter release at several types of synapses in the peripheral and central nervous systems (Chalazonitis 1996). NT-3 has been shown to have an important role in the overall development of enteric neurons, which are crucial for gut peristalsis (Chalazonitis 2004). Studies in rats have shown the potential of NT-3 in dorsal column axonal regeneration (Bradbury et al.). NT-3 was shown to protect neurons against amyloid-β toxicity (Lesne et al.). NT-3 has applications in neuronal differentiation protocols to generate β-tubulin III+ peripheral neurons from neural crest stem cells (Menendez et al.) and oligodendrocyte precursor cells from human embryonic stem (ES) and induced pluripotent stem (iPS) cells (Douvaras et al.).

The platelet-derived growth factor (PDGF) family has five heparin-binding members that assemble into four homodimers (PDGF-AA, PDGF-BB, PDGF-CC, and PDGF-DD) and one heterodimer (PDGF-AB; Fretto et al.; Li and Eriksson). PDGF signals through the receptor tyrosine kinases PDGFRα and PDGFRβ. It has been shown that PDGF-induced migration involves signaling pathways involving MEK/ERK, EGFR, Src, and PI3K/AKT (Kim et al.). Fibroblast growth factor 10 (FGF-10) is a member of the fibroblast growth factor (FGF) family predominantly expressed by mesenchymal fibroblasts during embryonic development (Emoto et al.; Igarashi et al.). Interleukin 17AF (IL-17AF) is a heterodimer that is composed of the interleukin 17A (IL-17A) and interleukin 17F (IL-17F) members of the IL-17 family of cytokines. IL-17AF is produced by T helper 17 cells (Th17) following interleukin 23 (IL-23) stimulation. N-Hexadecanoyl-DL-homoserine lactone (C16-HSL) is a small diffusible signaling molecule and is a member of N-acyl-homoserine lactone family. N-acylhomoserine lactones (AHL) are involved in quorum sensing, controlling gene expression, and cellular metabolism. C16-HSL is a lipophilic, long acyl side-chain bearing AHL, produced by the LuxI AHL synthase homolog SinI involved in quorum-sensing signaling in S. meliloti, a nitrogen-fixing bacterial symbiont of certain legumes. C16-HSL is the most abundant AHL produced by the proteobacterium R. capsulatus and activates genetic exchange between R. capsulatus cells. It tends to localize in relatively lipophilic cellular environments of bacteria and cannot diffuse freely through the cell membrane. The long-chain N-acylhomoserine lactones may be exported from cells by efflux pumps or may be transported between communicating cells by extracellular outer membrane vesicles.
