Angiotensin II is a potent vasopressor hormone and a primary regulator of aldosterone secretion that acts through at least two types of receptors, AGTR1 and AGTR2. It is an important effector controlling blood pressure and volume in the cardiovascular system and plays a major role in the development of the mammalian kidney and urinary tract. AGTR1, the type 1 receptor, is thought to mediate the major cardiovascular effects of angiotensin II and may play a role in the generation of reperfusion arrhythmias following restoration of blood flow to ischemic or infarcted myocardium. AGTR1 has recently been found to regulate the differentiation of bone marrow-derived monocyte lineage progenitors from hematopoietic stem cells, indicating the diversity of the roles of AGTR1.

AKT is a component of the PI-3 kinase pathway and is activated by phosphorylation at Ser 473 and Thr 308. AKT is a cytoplasmic protein also known as AKT1, Protein Kinase B (PKB) and rac (related to A and C kinases). AKT is a key regulator of many signal transduction pathways. AKT Exhibits tight control over cell proliferation and cell viability. Overexpression or inappropriate activation of AKT is noted in many types of cancer. AKT mediates many of the downstream events of PI 3-kinase (a lipid kinase activated by growth factors, cytokines and insulin). Dendritic cells (DCs) that control immune responses were recently found to capture and transport HIV from the mucosal area to remote lymph nodes, where DCs hand over HIV to CD4+ T lymphocytes. DCs also amplify the amount of virus and extend the duration of viral infectivity. Multiple strains of HIV-1, HIV-2 and SIV bind to DCs via DC-SIGN. ICAM-3 is the natural ligand for DC-SIGN. A DC-SIGN homologue (termed DC-SIGNR, L-SIGN, and DC-SIGN2) was identified recently. DC-SIGN forms a novel gene family with DC-SIGNR and many alternatively spliced isoforms of DC-SIGN and DC-SIGNR. The expression of DC-SIGN was found in mucosal tissues including placenta, small intestine, and rectum.

The translation of mRNA in eukaryotic cells is regulated by the presence of amino acids through multiple mechanisms. One such mechanism involves the evolutionarily conserved serine/threonine kinase TOR (Target of rapamycin, also known as mTOR), which regulates cell growth and cell cycle through its ability to integrate signals from nutrient levels and growth factors. One downstream target of TOR is the eukaryotic initiation factor 4E binding protein 1 (4E-BP1) whose phosphorylation prevents its association with eIF4E, preferentially stimulating translation of mRNAs containing long, highly structured 5'-UTRs. Rapamycin inhibits TOR resulting in reduced cell growth and reduced rates of cell cycle and cell proliferationl, at least in part by inhibiting the activity of TOR towards 4E-BP1.

DARC, also known as the Duffy antigen/chemokine receptor, is a seven-transmembrane protein homologous to the classical chemokine G-protein coupled receptors (GPCRs) with the exception of the motif required for G protein coupling. DARC can bind with high affinity several chemokines without transducing any signal, suggesting it may modulate the signals normally induced by these chemokines. Recently, DARC was found to interact with KAI1, a four transmembrane protein recently identified as a tumor metastasis suppressor protein. It is thought that tumor cells dislodged from the primary tumor and expressing KAI1 interact with DARC proteins expressed on vascular cells, transmitting a senescent signal to the tumor cells, while tumor cells that have lost KAI1 expression can proliferate and potentially give rise to metastases. At least three isoforms of DARC are known to exist.

CCAAT enhancer binding proteins (CEBPs) are a family of transcription factors that all contain a highly conserved, basic-leucine zipper domain at the C-terminus that is involved in dimerization and DNA binding. At least six members of the family have been isolated and characterized to date (CEBP alpha to CEBP zeta). CEBPD is a leucine zipper (LZ) DNA-binding protein that regulates gene expression in a variety of tissues including liver, adipose, lung and intestine. CEBPD is an important transcriptional activator in the regulation of genes involved in immune and inflammatory responses and has been reported to possess many tumor suppressor-like properties.
