Bacterial transformation, plasmid DNA isolation, and electrophoresis are all included in this comprehensive lab activity that can be completed in two 60 minute sessions. Students transform E. coli cells by inserting the plasmid pUC8 to confer antibiotic resistance on the cells and use an easy, safe mini-prep method to isolate the plasmid. Students then run a gel, comparing the isolated plasmid to a known sample to confirm that the plasmid is pUC8. The kit includes enough materials to run eight gels, a teacher’s guide, and student copymaster. An electrophoresis chamber, power supply, and 10 μL micropipets, required, are available separately.
